Multiphoton Phosphorescence Quenching Microscopy Reveals Kinetics of Tumor Oxygenation during Antiangiogenesis and Angiotensin Signaling Inhibition

John D. Martin et al.

Summary: This study reveals the spatial and temporal heterogeneity in tumor vessel function and oxygen levels, which can affect the response to therapeutic agents. The findings suggest that optimizing the dose and schedule of vascular and stromal normalizing strategies should take into account the dynamic changes in tumor vasculature.

Renin-angiotensin-aldosterone system inhibitors and survival in patients with hypertension treated with immune checkpoint inhibitors

Zsofia D. Drobni et al.

Summary: This retrospective study investigated the association between the use of renin-angiotensin-aldosterone system (RAAS) inhibitors and outcomes in patients treated with immune checkpoint inhibitors (ICIs). The results showed that hypertension patients who concomitantly took a RAAS inhibitor during ICI therapy had better overall survival. This benefit was particularly significant among patients with gastrointestinal and genitourinary cancers.

The Current Landscape of Immune Checkpoint Blockade in Hepatocellular Carcinoma: A Review

Matthias Pinter et al.

Summary: Recent advancements in the treatment of advanced hepatocellular carcinoma (HCC) have led to the approval of new systemic therapies such as angiogenesis inhibitors and immune checkpoint blockers, indicating important progress in the field. Strategies to target immunosuppression in tumors, such as vascular endothelial growth factor pathway inhibition and transforming growth factor-beta pathway inhibition, show promise in enhancing the efficacy of immunotherapy in HCC. Successes and failures of these treatments have provided insights into implementing novel systemic therapy in HCC and improving immunotherapy efficacy for this disease.

Normalizing the Microenvironment Overcomes Vessel Compression and Resistance to Nano-immunotherapy in Breast Cancer Lung Metastasis

Fotios Mpekris et al.

Summary: Nano-immunotherapy has shown potential in improving outcomes for triple negative breast cancer patients, but mechanisms of resistance need to be understood. By decompressing compressed blood vessels in lung metastases, perfusion can be restored and hypoxia alleviated, leading to more efficient delivery of nanomedicines and enhancement of anti-tumor immunity. Mechanotherapeutics in combination with nano-immunotherapy may offer synergistic effects on efficacy in breast cancer treatment.

Type I collagen deletion in αSMA+ myofibroblasts augments immune suppression and accelerates progression of pancreatic cancer

Yang Chen et al.

Summary: The stromal desmoplastic reaction in pancreatic ductal adenocarcinoma involves significant accumulation of type I collagen (Col1). Deletion of Col1 specifically in myofibroblasts accelerates PDAC progression by increasing Cxcl5 expression in cancer cells and promoting recruitment of myeloid-derived suppressor cells. Targeting CXCR2 and CCR2 may help restrain accelerated PDAC progression in the absence of stromal Col1.

STING agonist-based treatment promotes vascular normalization and tertiary lymphoid structure formation in the therapeutic melanoma microenvironment

Manoj Chelvanambi et al.

Summary: The activation of STING within the TME leads to slowed melanoma growth, increased production of antiangiogenic and TLS-inducing factors, as well as improved VN and enhanced tumor infiltration by immune cells. The delivery of low-dose STING agonist promotes a proinflammatory TME supportive of TLS formation and tumor growth control.

Overcoming microenvironmental resistance to PD-1 blockade in genetically engineered lung cancer models

Summary: In the treatment of nonsmall cell lung cancer, antiangiogenic therapy can slow tumor progression, but unlike in other cancers, the addition of PD-1 or PD-L1 antibodies does not provide benefits. Antiangiogenic treatment facilitates tumor infiltration by PD-1(+) T-regs, but targeting both tumor-associated macrophages types can improve treatment efficacy.

Bevacizumab improves tumor infiltration of mature dendritic cells and effector T-cells in triple-negative breast cancer patients

Yves Boucher et al.

Summary: The study showed that a single dose of bevacizumab can reduce the density of angiopoietin-2-positive vessels and increase the infiltration of immune cells in patients with primary triple-negative breast cancer, suggesting a potential rationale for combining bevacizumab with immune checkpoint blockers in neoadjuvant treatment to improve efficacy.

Endothelin Inhibition Potentiates Cancer Immunotherapy Revealing Mechanical Biomarkers Predictive of Response

Chrysovalantis Voutouri et al.

Summary: The effectiveness of immunotherapy relies on T cell migration to tumors and their ability to kill malignant cells. This study explores the potential of using the nonselective endothelin receptor blocker bosentan to normalize the tumor microenvironment and improve immunotherapy by reducing tumor stiffness and decompressing vessels. Ultrasound stiffness measurements show promise as a predictive biomarker for response to immune checkpoint inhibitors.
