CRISPR-based RNA-binding protein mapping in live cells

Baiwen Chen et al.

Summary: RBPs play essential roles in RNA metabolism and cell homeostasis, and a novel CRISPR-based method was developed to identify proteins binding to specific RNAs in live cells. The application of this method to U1 small nuclear RNA revealed 226 proteins associated with it.

CBRPP: a new RNA-centric method to study RNA-protein interactions

Yunfei Li et al.

Summary: RNA and protein can influence each other through physical interactions, and abnormal interactions can lead to cell dysfunctions and diseases. While many protein-centric methods exist for studying RNA-protein interactions, few robust RNA-centric methods are available. CBRPP is a new RNA-centric method that can identify proteins associated with a target RNA in native cellular context without manipulation of RNA-protein complexes in vitro.

Summary: The interaction between RNAs and proteins controls multiple cellular functions and specific knowledge is required to understand their interactions. Various techniques have been developed to study RNA-protein interactions including CLIP and RNA affinity purification. In vivo proximity labeling can be used to study RNA-protein interactions and analyze tagged molecules via mass spectrometry or RNA-Seq.

Summary: RNA molecules, including mRNAs encoding proteins and noncoding transcripts, function with the help of RNA binding proteins (RBPs). RNA proximity labelling (RPL), a method utilizing proximity-based biotinylation, has been developed to rapidly identify target RNA binding proteins in native cellular contexts. This technique has the potential to uncover novel RNA-protein interactions important for health and disease.

Summary: CRISPR-Cas13 is a versatile tool for controlling gene expression and cell fate at the RNA level, in addition to RNA interference, it can also modulate various aspects of RNA fate through RNA modifications. These toolkits have been applied in cell biology, disease, and imaging, but potential limitations such as off-target editing, low efficiency, and delivery restrictions exist.
