The ATM and ATR kinases regulate centrosome clustering and tumor recurrence by targeting KIFC1 phosphorylation

Guangjian Fan et al.

Summary: In this study, KIFC1 was found to be phosphorylated by ATM and ATR kinases under DNA damage conditions, promoting centrosome clustering and leading to tumor recurrence. Inhibition of KIFC1 phosphorylation can suppress tumor recurrence. The research suggests that centrosome clustering regulation may be a promising target for cancer therapy.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Overcoming Resistance to Tumor-Targeted and Immune-Targeted Therapies

Mihaela Aldea et al.

Summary: Resistance to anticancer therapies can be classified into primary and secondary resistance, primarily influenced by target dependency and cellular adaptability. Immunotherapy resistance is often driven by tumor-host-microenvironment interactions, while targeted therapy resistance is typically driven by alterations in targets.

Repositioning PARP inhibitors in the treatment of thoracic malignancies

Francesco Passiglia et al.

Summary: The evaluation of the HRR status is becoming an important predictive biomarker for PARP inhibition in various tumor types, particularly in thoracic malignancies. Clinical trials are currently being conducted to explore the efficacy of novel PARPi combinations in patients with lung cancer and malignant pleural mesothelioma, taking advantage of the deeper understanding of HRR-deficient tumors and the development of new targeted and immunotherapeutic agents.

Preoperative clinical and tumor genomic features associated with pathologic lymph node metastasis in clinical stage I and II lung adenocarcinoma

Raul Caso et al.

Summary: NGS was utilized to identify genetic factors associated with LN metastasis in LUAD patients, revealing higher rates of certain gene alterations in pN-positive tumors. Multivariable analysis identified independent factors associated with LN metastasis, providing guidance for preoperative therapy decisions.
