MIND diet and the risk of dementia: a population-based study

Assessment of neurovascular coupling and cortical spreading depression in mixed mouse models of atherosclerosis and Alzheimer's disease

Summary: Neurovascular coupling is key for brain health, but its disruption is linked to neurological diseases like dementia. This study examines cortical haemodynamics in mouse models of Alzheimer's disease and atherosclerosis, finding reduced blood volume, altered haemoglobin levels, and neuroinflammation in atherosclerosis, as well as increased amyloid-beta plaques in the hippocampus. Importantly, electrode insertion worsens cortical spreading depression and leads to prolonged hypoxia in diseased animals. Atherosclerosis can negatively affect neurovascular health, and cardiovascular comorbidities can worsen Alzheimer's-related plaques.

Reduced pericyte and tight junction coverage in old diabetic rats are associated with hyperglycemia-induced cerebrovascular pericyte dysfunction

Summary: This study demonstrates that the loss of contractile capability in pericytes in diabetes is associated with enhanced ROS and reduced ATP production. Enhanced advanced glycation end products (AGEs) in diabetes accompany with reduced pericyte and endothelial tight junction coverage in the cortical capillaries of old diabetic rats. These results suggest our previous findings that the impaired cerebral hemodynamics, BBB leakage, and cognitive impairments in old DM model are associated with hyperglycemia-induced cerebrovascular pericyte dysfunction.

However, there is still poor understanding of the molecular and cellular mechanisms causing these CBF deficits, and future research aimed at elucidating the interplay between different mechanisms and their role in AD patients could guide the development of therapies targeting CBF reductions.
