The transfection protocol was also successfully optimized for high mutation frequency in rapeseed gene editing.

Summary: This study focused on using the CRISPR-Cas9 genome editing system to target multiple homoeologs of BnSHP1 and BnSHP2 in Brassica napus to identify mutated lines with improved resistance to pod shattering. The results indicated that mutation of specific BnSHP1A09 homoeolog showed promise in controlling lignin contents, leading to decreased pod shattering. Mutations in BnSHP1A09/C04-B/A04 and BnSHP2A05/C04-A resulted in reduced lignified layer and separation layer, ultimately increasing the pod-shattering resistance index (SRI) in mutated lines compared to the wild type.
