A novel tissue-engineered bone graft composed of silicon-substituted calcium phosphate, autogenous fine particulate bone powder and BMSCs promotes posterolateral spinal fusion in rabbits

LiHuang Cui et al.

Summary: This study evaluated a novel tissue-engineered bone graft consisting of Si-CaP, AFPBP, and BMSCs in a rabbit posterolateral lumbar fusion model. The graft showed excellent bioactivity, biocompatibility, and biodegradability, reducing the amount of autogenous bone while promoting spinal fusion, providing a potential alternative to autogenous bone grafts.

A Review of Recent Developments in the Molecular Mechanisms of Bone Healing

Emerito Carlos Rodriguez-Merchan

Summary: This article reviews recent findings on the molecular mechanisms of bone healing and presents various biological alternatives for treating recalcitrant nonunions, such as using morin, BMP9, and leptin. It also discusses factors that impact bone regeneration, including inhibition of estrogen receptor signaling, smoking-induced inflammatory responses, and delayed fracture healing in diabetic animals due to BMP6 deficiency. The combination of bioceramics and expanded autologous human mesenchymal stem cells from bone marrow is highlighted as a promising alternative for treating nonunions that do not respond well to traditional treatments.

Biomimetic, Stiff, and Adhesive Periosteum with Osteogenic-Angiogenic Coupling Effect for Bone Regeneration

Yuhe Yang et al.

Summary: The study presents the development of a periosteum mimicking bone aid (PMBA) with enhanced mechanical strength, tissue adhesiveness, and sustained activation of nitric oxide-cyclic guanosine monophosphate (NO-cGMP) signaling pathway for accelerated bone regeneration. This concept involves considering both the initial material components and the degradation products in promoting physiological activities, aiming to inspire the design of high-performance bioscaffolds for bone and periosteum tissue engineering.

Biomineralization Directed by Prenucleated Calcium and Phosphorus Nanoclusters Improving Mechanical Properties and Osteogenic Potential of Antheraea pernyi Silk Fibroin-Based Artificial Periosteum

Yajun Shuai et al.

Summary: Using biomacromolecules as templates to control hydroxyapatite crystal nucleation and growth, an artificial periosteum is prepared by biomineralizing Antheraea pernyi fibroin (AF) membrane with prenucleated nanoclusters. This approach promotes osteogenic differentiation of MSCs, induces bone matrix protein formation, and enhances the mechanical properties of the AF membrane. The biomineralized AF membranes show good cytocompatibility, hemocompatibility, and histocompatibility in vitro and in vivo, making them a promising strategy in the field of bone tissue engineering.

Growth factor delivery using extracellular matrix-mimicking substrates for musculoskeletal tissue engineering and repair

Robert C. H. Gresham et al.

Summary: Therapeutic approaches for musculoskeletal tissue regeneration commonly use growth factors to influence nearby cells for migration, proliferation, or differentiation. However, challenges in clinical translation include poor control and high dosages leading to side effects. The extracellular matrix plays a key role in regulating growth factor presentation and retention, impacting tissue regeneration.
