Photothermal Conjugated Polymer Nanoparticles for Suppressing Breast Tumor Growth by Regulating TRPA1 Ion Channels

Summary: TRPA1 ion channels are overexpressed in breast cancer cells and enhance cellular resistance to oxidative stress through upregulating the anti-apoptotic pathway. Near-infrared light-responsive conjugated polymer nanoparticles are designed to regulate TRPA1 ion channels and promote apoptosis of breast cancer cells, reducing drug resistance and suppressing tumor growth.

Functional expression of the transient receptor potential ankyrin type 1 channel in pancreatic adenocarcinoma cells

Summary: This study reveals the endogenous expression of TRPA1 channels in human pancreatic adenocarcinoma cell lines and provides insights into the function of the TRPA1 protein, showing that different cell lines isolated from PDAC patients had varying levels of TRPA1 expression. Activation of TRPA1 by AITC in Panc-1 cells induced non-selective cationic currents, inhibited by the TRPA1 antagonist A-967079, and led to increased intracellular Ca2+ levels. siRNA-induced downregulation of TRPA1 enhanced cell migration, suggesting a role of ion channels independent of pore function, and TRPA1 activation altered cell cycle progression.

Comprehensive characterization of protein-protein interactions perturbed by disease mutations

Summary: Recent advancements in genomics and interactomics have allowed researchers to identify how disease mutations disrupt protein-protein interaction networks within human cells. Disease-associated germline variants and somatic missense mutations are significantly enriched in protein-protein interaction interfaces, and oncoPPIs identified in cancer analysis are correlated with patient survival and drug resistance/sensitivity. Experimental validation of oncoPPIs and their effects on tumor cell growth demonstrate the potential of using this interactome network framework for prioritizing therapeutic targets.

Dysfunctional TRPM8 signalling in the vascular response to environmental cold in ageing

Summary: Ageing is associated with increased vulnerability to environmental cold exposure, leading to impaired behavioural and vascular responses to skin local environmental cooling in aged mice. The decline in TRPM8 gene/protein expression in ageing mice suggests a reliance on remaining TRP receptor activity for cold-induced vascular response. Sympathetic-induced vasoconstriction is also reduced with aging, indicating a decline in the cold-induced vascular response necessary for maintaining body heat and health.
