Are lipid nanoparticles really superior? A holistic proof of concept study

Sabrina Wiemann et al.

Summary: Lipid nanoparticles are an effective carrier system for dermal drug delivery, offering increased chemical stability, dermal penetration, and skin carrying properties. This study demonstrated that lipid nanoparticles can combine all these benefits in one formulation, making them a superior choice compared to other formulation principles.

Assessing the Dermal Penetration Efficacy of Chemical Compounds with the Ex-Vivo Porcine Ear Model

Cornelia M. Keck et al.

Summary: The study showed that post-mortem changes in porcine ears can affect the penetration efficacy of chemical compounds. Regardless of the post-slaughter storage time, all types of ears are suitable for dermal penetration testing. SmartFilms and nanostructured lipid carriers were found to be superior strategies for efficient delivery of curcumin through the skin.

Hair follicle targeting with curcumin nanocrystals: Influence of the formulation properties on the penetration efficacy

Olga Pelikh et al.

Summary: Nanocrystals are a promising formulation for improved drug delivery, targeting the hair follicles and providing long-lasting and efficient dermal drug delivery. Different types of vehicles do not significantly affect the efficacy of nanocrystals' penetration into hair follicles, but they do impact passive dermal penetration. Gels containing humectants can enhance passive penetration effectiveness.

Non-ionic surfactants as innovative skin penetration enhancers: insight in the mechanism of interaction with simple 2D stratum corneum model system

Fabio Strati et al.

Summary: Transdermal drug delivery is a passive diffusion process where non-ionic surfactants can enhance drug penetration. This study focused on the interaction between surfactants and a lipidic matrix model system, comparing the effects of Polysorbate 80 with Sucrose monolaurate on disordering the SC monolayer.

Influence of Massage and Skin Hydration on Dermal Penetration Efficacy of Nile Red from Petroleum Jelly-An Unexpected Outcome

Vasudha Kaushik et al.

Summary: The study compared the influence of direct and indirect skin hydration as well as massage on the dermal penetration efficacy of active compounds, but the results obtained were contrary to expectations. It was found that Vaseline undergoes syneresis after topical application, leading to unexpected effects on dermal penetration.

Improved Dermal and Transdermal Delivery of Curcumin with SmartFilms and Nanocrystals

Ralph W. Eckert et al.

Summary: The study used smartFilm technology to improve the dermal penetration efficacy of curcumin, showing better results compared to nanosuspensions and bulk suspensions. Increasing the number of curcumin particles in the suspensions was found to increase the passive dermal penetration. These findings suggest a new dermal penetration mechanism for active compounds from nano-particulate drug delivery systems, which could lead to the development of topical drug products with improved penetration efficacy in the future.

Summary: This study aimed to develop a standard formulation for curcumin and a screening tool for fast discrimination of dermal penetration efficacy. The study utilized ethanolic curcumin solutions as standard formulations and an ex vivo porcine ear model with epifluorescence microscopy for evaluation. The results show that this skin penetration model is a versatile tool for determining dermal penetration efficacy and provides detailed information on the fate of chemical compounds after penetration.

Enhancing Permeation of Drug Molecules Across the Skin via Delivery in Nanocarriers: Novel Strategies for Effective Transdermal Applications

Yi-Qun Yu et al.

Summary: The transdermal route of administration offers benefits such as reduced toxicity and sustained drug delivery, but is limited by the stratum corneum barrier. Nanocarriers provide a promising solution to enhance drug penetration, while physical methods like iontophoresis and microneedles can further improve transdermal drug delivery efficacy.
