Optimization of diluents on the basis of SeDeM-ODT expert system for formulation development of ODTs of glimepiride

Amjad Khan et al.

Summary: The present study optimized the characteristics of diluents using the SeDeM-ODT expert system to mask poor characteristics of APIs and facilitate the development of a formulation using direct compression technique. The study demonstrated that the selected diluents based on the SeDeM-ODT expert system improved the flow and disintegration behavior of the powder blends, resulting in tablets with sufficient mechanical strength. This study provides a simplified and optimized approach for formulation development.

Use of machine learning in prediction of granule particle size distribution and tablet tensile strength in commercial pharmaceutical manufacturing

Eero Maki-Lohiluoma et al.

Summary: In this study, predictive machine learning models were used to predict Particle Size Distribution (PSD) and Tensile Strength (TS) for a commercial tablet product, aiming to indicate product quality. Feature importance and partial dependence were utilized to evaluate parameter importance and impact on predicted TS. The study demonstrates the ability of machine learning models to enhance product-related knowledge and bring additional value for commercial products.

Formulation and Quality Control of Orally Disintegrating Tablets (ODTs): Recent Advances and Perspectives

Mohammadali Poursharifi Ghourichay et al.

Summary: Orally disintegrating tablets (ODTs) are a type of dosage form that rapidly disintegrates or dissolves in the oral cavity without using water, improving drug bioavailability. ODTs are commonly used in patients with swallowing challenges, children, and cases where rapid onset of action is needed.

An insight into predictive parameters of tablet capping by machine learning and multivariate tools

Shubhajit Paul et al.

Summary: The study characterized 26 diverse formulations under commercial tableting conditions to identify key tablet properties influencing capping propensity, and established a predictive model using machine learning and multivariate tools based on threshold properties. The study found that both compaction parameters and material properties strongly dictate the capping propensity of a tablet, with the ratio of elastic modulus and its variation with porosity being notable quantitative metrics of capping occurrence.

Design of an orally disintegrating tablet formulation containing metoprolol tartrate in the context of quality by design approach

Gizem Yegen et al.

Summary: Orally Disintegrating Tablets (ODTs) are solid dosage forms that rapidly disintegrate or dissolve in the mouth upon contact with saliva. Quality by Design (QbD) approach, with the help of artificial intelligence programs, was used to develop an ODT formulation containing Metoprolol tartrate, aiming to understand the relationship between critical parameters and quality attributes and to optimize the formulation for compliance with pharmacopoeia limits. Adopting the QbD approach and using artificial intelligence programs have increased the efficiency of the formulation development process with better understanding of the product and process.
