Integrated single-cell and bulk RNA sequencing analysis identifies a cancer associated fibroblast-related signature for predicting prognosis and therapeutic responses in colorectal cancer

Hang Zheng et al.

Summary: The study established a nine-gene prognostic CAF-related signature for CRC, which was validated in multiple cohorts. Patients with higher CAF risk scores showed adverse prognosis and higher CAF infiltrations. The potential strategy of combining chemotherapy with anti-CAF therapy was suggested to improve the efficacy of T-cell based immunotherapies.

Differential pre-malignant programs and microenvironment chart distinct paths to malignancy in human colorectal polyps

Bob Chen et al.

Summary: Colorectal cancers originate from different precursor polyps, with factors related to stem cell expansion and cell differentiation playing key roles in tumor development; changes in the immune environment can also lead to mutations in tumor cells.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Tumor Microenvironment Profiles Reveal Distinct Therapy-Oriented Proteogenomic Characteristics in Colorectal Cancer

Nan Wang et al.

Summary: The advancements in immunotherapy for treating colorectal cancer have shown significant immune infiltration at tumor regions associated with immunotherapy treatment. The correlation between protein and mRNA levels within the same selected regions of patient samples suggests an overall concordance between transcriptional and translational levels. An elevated expression of PD-L1 at both protein and mRNA levels was discovered in the tumor compartment of immunotherapy-treated patients compared with chemo-treated patients.

The Adaptive Immune Landscape of the Colorectal Adenoma-Carcinoma Sequence

Joao Augusto Freitas et al.

Summary: The study found that the sporadic colorectal ACS is characterized by a gradual decrease in immune cell counts, an increase in TMB and MHC-I expression, and lower PD-L1 expression. Similar patterns were observed in FAP lesions, with the exception of an increase in TMB along the ACS and lower Foxp3+ T cell counts in FAP LGD lesions compared to sporadic ones. PD-L1 expression decreased earlier in FAP lesions than in sporadic ones.
