Summary: This study demonstrated that CAFs-derived exosomes confer cisplatin resistance to NSCLC cells by transferring miRNA-130a, with PUM2 identified as a critical factor in packaging miRNA-130a into the exosomes. The results suggest that CAFs-derived exosomal miRNA-130a may be a potential therapeutic target for cisplatin resistance in NSCLC.

Downregulated exosomal microRNA-148b-3p in cancer associated fibroblasts enhance chemosensitivity of bladder cancer cells by downregulating the Wnt/β-catenin pathway and upregulating PTEN

Guang Shan et al.

Summary: Exosomes derived from cancer-associated fibroblasts (CAFs) play a crucial role in promoting the metastasis, EMT, and drug resistance of bladder cancer cells through delivering miR-148b-3p. PTEN has been identified as a direct target of miR-148b-3p, and its overexpression can inhibit EMT, metastasis, and chemoresistance in bladder cancer cells. Downregulation of miR-148b-3p in CAF-derived exosomes may offer potential therapeutic opportunities for bladder cancer treatment by inhibiting the Wnt/beta-catenin pathway and promoting PTEN expression.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Chitinase 3-like-1 and fibronectin in the cargo of extracellular vesicles shed by human macrophages influence pancreatic cancer cellular response to gemcitabine

Cristina P. R. Xavier et al.

Summary: Research has shown that large EVs released by macrophages contain proteins CHI3L1 and FN1, whose overexpression can lead to increased resistance of PDAC cells to gemcitabine. Partial reversal of gemcitabine resistance can be achieved by inhibiting CHI3L1 and FN1, providing potential targets for pharmacological intervention in PDAC treatment.

Cancer-associated fibroblast-derived exosomal microRNA-24-3p enhances colon cancer cell resistance to MTX by down-regulating CDX2/HEPH axis

Hong-Wei Zhang et al.

Summary: In colon cancer, high expression of miR-24-3p and low expression of HEPH and CDX2 were observed in tissues and cells. CDX2, targeted by miR-24-3p, could up-regulate HEPH. Inhibition of CAF-derived exosomal miR-24-3p decreased cell viability and colony formation, and increased apoptosis, indicating a role in promoting chemotherapy resistance in colon cancer.

The cross-talk between tumor cells and activated fibroblasts mediated by lactate/BDNF/TrkB signaling promotes acquired resistance to anlotinib in human gastric cancer

Zhijian Jin et al.

Summary: In this study, anlotinib inhibited gastric cancer (GC) cell growth by inducing apoptosis and G2/M phase arrest in a dose- and time-dependent manner. The interaction between epithelial and stromal cells in the tumor microenvironment affects the response of GC cells to anlotinib. Secreted lactate from GC cells activates cancer-associated fibroblasts (CAFs) to produce BDNF in a NF-kappa B-dependent manner, reducing the sensitivity of GC cells to anlotinib. Targeting the BDNF-TrkB pathway increased the sensitivity of GC cells to anlotinib in a human patient-derived organoid model.

RAB27A-dependent release of exosomes by liver cancer stem cells induces Nanog expression in their differentiated progenies and confers regorafenib resistance

Perspectives on Hypoxia Signaling in Tumor Stroma

Yuqing Zhang et al.

Summary: Hypoxia is a common feature of solid tumors that promotes tumor progression and metastasis. Hypoxia, mainly mediated by the HIF family, leads to tumor cell migration, proliferation, metabolic changes, and therapy resistance. Additionally, hypoxia affects various cellular and non-cellular compartments in the tumor stroma, including disordered tumor vasculature and ECM homeostasis. Hypoxia also has complex and sometimes contradictory effects on immune cell function, contributing to an immunosuppressive environment.

Summary: Hypoxia exacerbates cisplatin resistance in NSCLC cells by increasing the expression of PKM2 in exosomes secreted by hypoxic cisplatin-resistance cells. Exosomal PKM2 transmits cisplatin resistance to sensitive NSCLC cells by promoting glycolysis and inhibiting apoptosis. Additionally, exosomal PKM2 remodels CAFs to create an acidic microenvironment that promotes NSCLC cell proliferation and cisplatin resistance.

Extracellular vesicles derived from hypoxic glioma stem-like cells confer temozolomide resistance on glioblastoma by delivering miR-30b-3p

Summary: The study found that EVs from hypoxic GSCs exerted a greater impact on GBM chemoresistance, with significantly upregulated miR-30b-3p. HIF1 alpha and STAT3 transcriptionally induced miR-30b-3p expression, while hnRNPA2B1 facilitated its transfer into EVs, targeting RHOB and reducing apoptosis while promoting proliferation in vitro and in vivo. The findings suggest that targeting EV-miR-30b-3p could be a potential treatment strategy for GBM.
