EMT, cancer stem cells and autophagy; The three main axes of metastasis

Ghader Babaei et al.

Summary: Epithelial-mesenchymal transition (EMT) and Cancer stem-like cells (CSCs) are major factors contributing to cancer cell metastasis. Autophagy has recently gained attention, playing a dual role in cancer cell development depending on cancer type, tissue, and stage. Studies show different signaling pathways are involved in cancer stem cells, autophagy, and EMT. Investigating effective signaling pathways in all three processes could help determine appropriate therapeutic goals.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

The novel long noncoding RNA AU021063, induced by IL-6/Arid5a signaling, exacerbates breast cancer invasion and metastasis by stabilizing Trib3 and activating the Mek/Erk pathway

Kishan Kumar Nyati et al.

Summary: The study revealed that IL-6 induces the expression of AU021063 in breast cancer through Arid5a, and AU021063 further promotes breast cancer metastasis by stabilizing Trib3 and activating the Mek/Erk signaling pathway.

Microfluidic System to Analyze the Effects of Interleukin 6 on Lymphatic Breast Cancer Metastasis

Hyeon-Yeol Cho et al.

Summary: Research has shown that in a three-channel microfluidic system mimicking lymphatic metastasis pathways, the inflammatory cytokine IL-6 can induce epithelial-mesenchymal transition and enhance tissue invasion of breast cancer cells, as well as promote human vein endothelial cell growth towards lymphatic channels.

Targeting the IL-6/STAT3 Signalling Cascade to Reverse Tamoxifen Resistance in Estrogen Receptor Positive Breast Cancer

Ho Tsoi et al.

Summary: This study identifies the molecular mechanisms through which BQ323636.1 enhances IL-6/STAT3 signaling, promoting tamoxifen resistance in ER+ breast cancer. High IL-6R expression is significantly associated with tamoxifen resistance, and targeting IL-6R with Tocilizumab can reverse this resistance. Analysis of clinical samples confirmed the correlation between IL-6R expression, BQ expression, and tamoxifen resistance in primary breast cancer. High IL-6R expression is linked to poorer survival outcomes in ER+ breast cancer.

Summary: The study demonstrates that glyceryl trinitrate (GTN) inhibits the migration and invasion of triple-negative breast cancer cells by supressing the activation of the JAK2/STAT3 signaling pathway. This suggests a potential application of GTN in the treatment of breast cancer, providing a new research direction.

Points to consider for the treatment of immune-mediated inflammatory diseases with Janus kinase inhibitors: a consensus statement

Peter Nash et al.

Summary: The use of Janus kinase inhibitors (JAKi) in immune-mediated inflammatory diseases has been extensively reviewed to provide guidance on efficacy and safety. A consensus statement was formed by an expert committee, evaluating 26 key points in six areas, with high levels of agreement and evidence-based recommendations. This consensus offers valuable insights for the practical management of JAKi in clinical practice.
