Quality of life from cytoreductive surgery in advanced ovarian cancer: Investigating the association between disease burden and surgical complexity in the international, prospective, SOCQER-2 cohort study

Sudha Sundar et al.

Summary: This study aimed to investigate the quality of life (QoL) after surgical resection for advanced ovarian cancer and explore the association with surgical complexity and disease burden. The results showed that increasing surgical complexity did not lead to a decline in QoL. For patients with advanced ovarian cancer, higher surgical complexity was associated with greater improvement in postoperative QoL.

Decellularized normal and cancer tissues as tools for cancer research

E. Gentilin et al.

Summary: It is widely accepted that cancer initiation involves genetic modifications, but a single genetic alteration is not enough for cancer development. Tumors are now seen as a mix of cells and extracellular matrix (ECM) forming a unique microenvironment. Research has shown that interactions between tumor cells and the cancer microenvironment play a crucial role in cancer progression and therapy response.

Summary: This study developed a novel ex vivo explant model that maintains the 3D architecture and microenvironmental cues of human pancreatic ductal adenocarcinoma (PDAC). The model showed advantages over standard organoids, retaining functional multi-cellular stroma and fibrosis, providing unprecedented opportunities for studying PDAC biology and personalized treatment.

Engineering a Vascularized 3D Hybrid System to Model Tumor-Stroma Interactions in Breast Cancer

Filipa C. Teixeira et al.

Summary: The stromal microenvironment of breast tumors plays a crucial role in tumor development and metastasis, with tumor angiogenesis requiring cooperation between various cell types and extracellular matrix. The developed hybrid alginate-based 3D system allows for the simulation and support of epithelial morphogenesis and endothelial tubulogenesis in breast tumors.

New Predictive Biomarkers for Ovarian Cancer

Ghofraan Abdulsalam Atallah et al.

Summary: Ovarian cancer is often diagnosed in advanced stages due to lack of early symptoms. Surgical debulking and chemotherapy are mainstays of treatment, but chemoresistance is a common issue. Therefore, there is a critical need for identifying new biomarkers to improve diagnosis and prognosis for ovarian cancer.

Generation of a three-dimensional in vitro ovarian cancer co-culture model for drug screening assays

Larissa Bueno Tofani et al.

Summary: The research on 3D co-cultures of ovarian cancer cells with mesenchymal cells and fibroblasts demonstrated higher spheroid regularity, upregulated gene expression related to tumorigenesis, angiogenesis, and metastases, and increased chemoresistance to Paclitaxel compared to 2D culture. The presence of mesenchymal cells and fibroblasts in the 3D co-culture model better recapitulated the in vivo tumor microenvironment and provided a more accurate evaluation of drug efficacy for ovarian cancer therapy.

Genetically Defined, Syngeneic Organoid Platform for Developing Combination Therapies for Ovarian Cancer

Shuang Zhang et al.

Summary: Genetically informed, immunocompetent tumor models are crucial for evaluating conventional, targeted, and immune therapies. By engineering mouse fallopian tube organoids, researchers were able to study the effects of specific mutations found in high-grade serous tubo-ovarian cancer (HGSC) models, leading to the development of effective combination treatments for different genetic subgroups of HGSC. These findings highlight the importance of genotype-informed, syngeneic organoid models for understanding tumor biology and therapeutic responses.

Summary: This study demonstrates that the efficacy of immune therapies in high-grade serous ovarian cancer is driven by inducing state changes in NK cells and specific subsets of T cells. The results suggest that immune therapies, such as BRD1 inhibitors, capable of inducing such cellular state changes may have increased effectiveness in treating HGSC.
