Anti-Tumor Role of CAMK2B in Remodeling the Stromal Microenvironment and Inhibiting Proliferation in Papillary Renal Cell Carcinoma

Qingan Jia et al.

Summary: In this study, we identified the core effector molecule CAMK2B as playing a crucial role in regulating the tumor microenvironment (TME) in kidney renal papillary cell carcinoma (KIRP). High stromal scores were associated with poor prognosis in KIRP. Experimental validation showed that CAMK2B promotes TME remodeling and inhibits proliferation in KIRP. Downstream effectors of CAMK2B in KIRP were identified as vascular endothelial growth factor (VEGF), transforming growth factor (TGF)beta, and close homolog of L1 (CHL1).

Membrane Damage during Ferroptosis Is Caused by Oxidation of Phospholipids Catalyzed by the Oxidoreductases POR and CYB5R1

Bo Yan et al.

Summary: Ferroptosis is a form of necrotic cell death caused by iron-dependent peroxidation of polyunsaturated phospholipids on cell membranes, which is carried out by oxidoreductases transferring electrons to oxygen to generate hydrogen peroxide for lipid peroxidation. This process needs to be counteracted by antioxidant systems to prevent cell death.

Summary: Cell suicide pathways, known as regulated cell death (RCD), are crucial in organismal development, homeostasis, and pathogenesis. This review discusses key RCD modalities such as apoptosis, entosis, necroptosis, pyroptosis, and ferroptosis, and how they serve as a defense mechanism against cancer while also being exploited in oncogenesis. Therapeutic agents that activate RCD are outlined, along with novel strategies for tumor elimination, highlighting the critical importance of RCD in combating cancer.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

A novel lncRNA-miRNA-mRNA competing endogenous RNA regulatory network in lung adenocarcinoma and kidney renal papillary cell carcinoma

Qiwei Zhou et al.

Summary: Through various analysis methods, it was found that GPRIN1 is up-regulated in KIRP and LUAD, and is associated with poor prognosis. LINC00894/MMP25-AS1/SNHG1/LINC02298/MIR193BHG-miR-140-3p is likely the most promising upstream regulatory pathway for GPRIN1. Upregulation of LINC00894/MMP25-AS1/SNHG1/LINC02298/MIR193BHG and downregulation of miR-140-3p are relevant to poor outcomes of KIRP and LUAD. The expression of GPRIN1 is significantly correlated with tumor immune cell infiltration, immune cell biomarkers, and immune checkpoints.
