Non-invasive evaluation of response to obeticholic acid in patients with NASH: Results from the REGENERATE study

Mary E. Rinella et al.

Summary: Non-invasive tests (NITs) have potential applications in evaluating fibrosis and treatment response in patients with non-alcoholic steatohepatitis (NASH). Treatment with obeticholic acid (OCA) can lead to reduced levels of alanine aminotransferase, aspartate aminotransferase, and gamma-glutamyltransferase, as well as improvements in Fibrosis-4, FibroTest, FibroMeter, and FibroScan-AST scores.

Randomised clinical trial: Semaglutide versus placebo reduced liver steatosis but not liver stiffness in subjects with non-alcoholic fatty liver disease assessed by magnetic resonance imaging

Anne Flint et al.

Summary: In patients with NAFLD, the change in liver stiffness between semaglutide and placebo was not significantly different, but semaglutide significantly reduced liver fat content compared to placebo. Additionally, semaglutide also led to decreases in liver enzymes, body weight, and HbA(1c) levels.

Hepatic stellate cell senescence in liver fibrosis: Characteristics, mechanisms and perspectives

Mengfan Zhang et al.

Summary: Myofibroblasts are crucial in fibrogenesis, with hepatic stellate cells being their main precursors. Senescent hepatic stellate cells have been identified in liver fibrosis, displaying reduced collagen production and proliferation. The introduction of senolytics as a treatment may help remove senescent cells and mitigate the adverse effects of their persistence in liver fibrosis.

Summary: In a randomized trial, the pan-PPAR agonist Lanifibranor showed significant improvement in histologic features of NASH at a dose of 1200 mg. However, adverse events such as weight gain, anemia, peripheral edema, diarrhea, and nausea were more common with Lanifibranor compared to placebo.

Single-cell and bulk transcriptomics of the liver reveals potential targets of NASH with fibrosis

Zhong-Yi Wang et al.

Summary: This study analyzed 17,810 non-parenchymal cells from six healthy human livers, identifying potential intercellular signaling axes and master regulators involved in HSC activation during fibrogenesis. It also identified 61 liver fibrosis-associated genes that may serve as drug target candidates for NASH-related fibrosis. Regulon analysis highlighted CREB3L1 as a master regulator of many of these genes, providing insight into cell-cell interactions and master regulators underlying HSC activation in liver fibrosis.

Summary: This article summarizes the main components and mechanisms involved in the progression of liver fibrosis, with a special focus on the metabolic regulation of key effectors such as hepatic stellate cells (HSCs). It also discusses the role of RNA-binding proteins (RBPs) in the post-transcriptional regulation that affects the progression of fibrosis and chronic liver diseases (CLD).

Mechanisms of Cellular Senescence: Cell Cycle Arrest and Senescence Associated Secretory Phenotype

Ruchi Kumari et al.

Summary: Cellular senescence is a stable cell cycle arrest triggered in response to various stimuli, characterized by dynamic changes in senescent cells. It has both positive effects on tissue repair and anti-cancer mechanisms, but can also negatively impact organismal health. Multiple pathways, including p53/p21 and p16/pRB, play central roles in regulating this process.

The Emergence of Senescent Surface Biomarkers as Senotherapeutic Targets

Martina Rossi et al.

Summary: Senetherapeutics are emerging to suppress the detrimental effects of senescence by targeting either senomorphics or senolytics. The main goal is to suppress the proinflammatory senescence-associated secretory phenotype and promote immune recognition and elimination of senescent cells. One attractive approach is targeting specific molecules or proteins on the surface of senescent cells for senolysis.
