Current and New Next-Generation Sequencing Approaches to Study Mitochondrial DNA

Summary: Traditional techniques have been used for studying mtDNA defects, but NGS as a new method has begun to be applied. A study validated that PCR-based library preparations are suitable for generating full accurate mtDNA sequences and accurately assessing heteroplasmy for single point mutations, while showing that a new PCR-free method performed best in detecting break positions and heteroplasmy of single large deletions.
