Desmosomal Cadherins in Health and Disease

Marihan Hegazy et al.

Summary: Desmosomal cadherins are a recently evolved adhesive substance that forms the core of desmosomes, which are crucial for tissue integrity. They not only resist mechanical stress, but also promote tissue differentiation and morphogenesis. Interfering with desmosomal cadherin function can lead to various diseases. Our understanding of their regulation and function is still limited, but new directions for combating desmosome-related diseases are emerging.

In Translation: FcRn across the Therapeutic Spectrum

Timothy Qi et al.

Summary: This review examines the biological and thermodynamic properties of FcRn and recent advancements in the field, including Fab determinants of FcRn affinity, transgenic preclinical models, and FcRn targeting for immune-complex clearance. It also evaluates the potential of therapeutic antibodies for SARS-CoV-2 to saturate FcRn-mediated recycling. The importance of endosomal transit parameters in the quantitative relationship between in vivo IgG persistence and in vitro FcRn binding is emphasized through modeling and simulation studies.

Autoantibody-Specific Signalling in Pemphigus

Thomas Schmitt et al.

Summary: Pemphigus is a severe autoimmune disease that impairs barrier functions of the epidermis and mucosa by targeting desmosomal adhesion molecules Dsg1 and Dsg3. Therapy for pemphigus is focusing on specific suppression of autoantibody formation, but additional treatment options to stabilize desmosomes during acute phase and relapses may be beneficial. Autoantibodies interfere with desmosome adhesion through inhibiting Dsg adhesion and engaging signaling pathways, leading to different clinical pemphigus phenotypes.
