Randomized Trial of Ruxolitinib in Antiretroviral-Treated Adults With Human Immunodeficiency Virus

Vincent C. Marconi et al.

Summary: This study confirms the safety and efficacy of Ruxolitinib for HIV-infected individuals on ART. While there was no significant reduction in plasma IL-6 levels, Ruxolitinib significantly decreased markers of immune activation and cell survival.

Impact of HIV infection on aging and immune status

Jose-Ramon Blanco et al.

Summary: Despite the advancements in antiretroviral therapy, individuals living with HIV still face elevated immune activation and inflammation which contribute to morbidity and mortality. Current interventions targeting specific mediators have not been effective in significantly reducing immune activation or its consequences, highlighting the need for more research to identify potential therapeutic targets to improve the life expectancy of HIV-infected individuals.

Programmed Cell Senescence in the Mouse Developing Spinal Cord and Notochord

Jorge Antolio Dominguez-Bautista et al.

Summary: The study found discrete areas with high senescence-associated beta galactosidase in the developing mouse spinal cord and notochord, and treatment with the senolytic ABT-263 decreased the number of motoneurons, supporting their senescent phenotype.

Senolytics and senomorphics: Natural and synthetic therapeutics in the treatment of aging and chronic diseases

Sofia M. Lagoumtzi et al.

Summary: Cellular senescence is a complex process influenced by genetic, epigenetic, and environmental factors, potentially contributing to aging and chronic diseases. Senescent cells exhibit a specific senescence-associated secretory phenotype, which, when amassed, leads to chronic inflammation-induced aging, creating a vicious cycle of reduced cell clearance. Targeting senescent cells and their secretory phenotype is seen as a potential therapeutic strategy for age-related pathologies.

Overcoming the senescence-associated secretory phenotype (SASP): a complex mechanism of resistance in the treatment of cancer

Cecilia R. Chambers et al.

Summary: Senescence is a cellular state where cells halt the cell cycle in response to stress, with senescent cancer cells and adjacent cells potentially remaining metabolically active and secreting pro-inflammatory factors known as the SASP. The SASP plays a critical role in tumorigenesis, affecting various processes and treatment efficacy. Future research on therapy-induced senescence and pharmacological manipulation may provide new avenues for cancer therapeutics.

Lesley R. de Armas et al.

Summary: The study used the dual reporter virus HIVGKO to investigate latency establishment and maintenance in CD4(+) T cells, and analyzed latently infected cells using single cell technologies. The research found that JAK1/2 inhibitors could reduce HIV infection events and block HIV reactivation from latent cells.

Treatment with the BCL-2/BCL-xL inhibitor senolytic drug ABT263/Navitoclax improves functional hyperemia in aged mice

Stefano Tarantini et al.

Summary: The present study found that NVC responses were significantly impaired in aged mice, and treatment with ABT263/Navitoclax improved NVC response and hippocampal-encoded functions of learning and memory. The findings suggest that senescent cells in the aged brain may contribute to age-related neurovascular uncoupling, exacerbating cognitive decline. The neurovascular protective effects of ABT263/Navitoclax treatment highlight the potential of senolytic treatments as effective interventions in patients at risk for vascular cognitive impairment.
