Cytokine release syndrome and associated neurotoxicity in cancer immunotherapy

Emma C. Morris et al.

Summary: CAR T cell therapy has revolutionized the field of cancer treatment, but significant toxicities can occur in up to one-third of patients. The most common toxicities include cytokine release syndrome and immune effector cell-associated neurotoxicity syndrome. Understanding their pathophysiology is crucial for developing novel therapeutics for prevention and management.

Taming the beast: CRS and ICANS after CAR T-cell therapy for ALL

Vipul Sheth et al.

Summary: Treatment with CD19 or CD22-targeted CAR-T cells shows promising efficacy in refractory or relapsed ALL patients, but is associated with significant toxicities, including CRS and ICANS. Understanding predictors of toxicity severity and developing strategies to enhance the safety of CAR T-cell therapy are crucial in improving outcomes for ALL patients.

CD70-specific CAR T cells have potent activity against acute myeloid leukemia without HSC toxicity

Tim Sauer et al.

Summary: CD70 is a promising target antigen for AML treatment, and CD70-CAR T cells demonstrate good proliferation and antitumor activity in vitro and in vivo for CD70-positive AML patients, showing potential as a new treatment strategy.

Transient rest restores functionality in exhausted CAR-T cells through epigenetic remodeling

Evan W. Weber et al.

Summary: This study showed that transient cessation of chimeric antigen receptor (CAR) signaling can enhance the efficacy of CAR-T cells, reactivating their anti-tumor functions, and challenging the notion that exhaustion is an epigenetically fixed state.

SynNotch CAR circuits enhance solid tumor recognition and promote persistent antitumor activity in mouse models

Axel Hyrenius-Wittsten et al.

Summary: The synthetic Notch (synNotch) CAR circuits improve the specificity and persistent antitumor activity of therapeutic T cells in solid tumors by preventing CAR-mediated tonic signaling, allowing T cells to maintain long-lived memory and non-exhausted phenotype. ALPPL2 is established as a clinically viable cell therapy target for multiple solid tumors, and synNotch CAR T cells demonstrate multifaceted therapeutic benefits.

Effective Killing of Acute Myeloid Leukemia by TIM-3 Targeted Chimeric Antigen Receptor T Cells

Wen-Hsin Sandy Lee et al.

Summary: AML relapse is mainly caused by MRD, especially therapy-resistant LSCs. This study proposed a new treatment strategy using anti-TIM-3 CAR T cells to target LSCs, showing promising results in eliminating AML cells and primary LSCs in patients. By eradicating LSCs with anti-TIM-3 CAR T-cell therapy, it may improve the clinical outcomes of AML patients.

Summary: CAR T cell therapies targeting haematological malignancies show promising efficacy, with CD28- and 4-1BB-derived costimulatory domains demonstrating potentially different cellular and clinical effects. Further research is needed to compare their effectiveness and toxicity profiles.

NOT-Gated CD93 CAR T Cells Effectively Target AML with Minimized Endothelial Cross-Reactivity

Rebecca M. Richards et al.

Summary: CD93 CAR T cells show promising efficacy in eliminating AML while sparing HSPCs, but exhibit on-target, off-tumor toxicity to endothelial cells. Co-expression of other AML targets on endothelial cells is identified, and a novel NOT-gated strategy is introduced to mitigate endothelial toxicity, along with the use of high-dimensional transcriptomic profiling for rational design of combinatorial immunotherapies.
