Breast Cancer Screening Strategies for Women With ATM, CHEK2, and PALB2 Pathogenic Variants A Comparative Modeling Analysis

Kathryn P. Lowry et al.

Summary: Annual MRI screening starting at 30 to 35 years followed by annual MRI and mammography at 40 years may reduce breast cancer mortality by more than 50% for women with ATM, CHEK2, and PALB2 pathogenic variants. In the setting of MRI screening, mammography prior to 40 years may offer little additional benefit.

Rapid Genetic Testing forBRCA1andBRCA2Mutations at the Time of Breast Cancer Diagnosis: An Observational Study

Kelly A. Metcalfe et al.

Summary: The study showed that providing rapid genetic testing at the time of breast cancer diagnosis helps women make treatment decisions, with the majority of BRCA mutation carriers choosing to undergo bilateral mastectomy.

A Population-Based Study of Genes Previously Implicated in Breast Cancer

Chunling Hu et al.

Summary: This study provides population-based estimates of the prevalence and risk of breast cancer associated with pathogenic variants in known cancer-predisposition genes. The results highlight the varying levels of risk associated with different genes, such as high risk for BRCA1 and BRCA2 and moderate risk for PALB2, as well as associations with specific subtypes of breast cancer for genes like RAD51C and RAD51D.

Breast Cancer Risk Genes - Association Analysis in More than 113,000 Women

Leila Dorling et al.

Summary: This study sequenced samples from 60,466 women with breast cancer and 53,461 controls using a panel of 34 putative susceptibility genes, identifying associations between protein-truncating variants in certain genes and breast cancer risk. The results provide important information for genetic counseling and define genes that are most clinically useful for predicting breast cancer risk.

Cost-Effectiveness of Targeted Genetic Testing for Breast and Ovarian Cancer: A Systematic Review

Andreas Koldehoff et al.

Summary: This study evaluated the results and quality of cost-effectiveness modeling studies on targeted genetic-based screen-and-treat strategies for preventing breast and ovarian cancer. The findings suggest that targeted BRCA or multiple gene screening is likely to be cost-effective. Methodological variations could be decreased by developing a reference model for future validation of cost-effectiveness models.

Frequency of Contralateral Prophylactic Mastectomy in Breast Cancer Patients with a Negative BRCA1 and BRCA2 Rapid Genetic Test Result

Kelly A. Metcalfe et al.

Summary: This study found that women use risk assessment and negative BRCA1/BRCA2 genetic test results to guide their surgical decisions. The uptake of CPM decreased for women who were initially considering CPM before genetic testing after receiving a negative BRCA1/BRCA2 genetic test result. The study also showed that the mother's breast cancer status influenced the decision to undergo CPM.

Implementation of Multigene Germline and Parallel Somatic Genetic Testing in Epithelial Ovarian Cancer: SIGNPOST Study

Dhivya Chandrasekaran et al.

Summary: Multigene testing in ovarian cancer, especially after counselling by multidisciplinary clinical teams, shows a high uptake rate of 97% and identifies a significant proportion of patients with pathogenic variants such as BRCA1/BRCA2, RAD51C/RAD51D/BRIP1. Prospective parallel somatic and germline panel testing is recommended to maximize variant identification in cancer patients.
