Intratumoral Foxp3+RORγt+ T cell infiltration determines poor prognosis and immunoevasive contexture in gastric cancer patients

Summary: High infiltration of Foxp3(+)ROR gamma t(+) T cells predicts poor overall survival and therapeutic response in gastric cancer. These cells are associated with impaired effective function of CD8(+) T cells and can more precisely predict survival outcomes and chemotherapeutic responsiveness. The novel co-evaluation system might be useful for prognosis prediction and appropriate treatment in gastric cancer.

Summary: The study found that neoadjuvant chemotherapy increases infiltration of cytotoxic CD8(+) T cells and CD20(+) B cells, and induces phenotypic changes in CD8(+) and CD4(+) memory T cells. Although neoadjuvant chemotherapy did not affect T-cell receptor clonality or tumor mutational burden, it promoted antitumor immunity through T and B cell recruitment in the immune microenvironment.

Global analysis of shared T cell specificities in human non-small cell lung cancer enables HLA inference and antigen discovery

Summary: Through analyzing a large number of T cell receptor sequences, we identified T cell receptors with shared antigen specificity in a subset of non-small cell lung cancer patients and identified some specificity groups enriched in tumors. Using a yeast peptide-HLA display library, we confirmed the antigenic epitopes of one tumor-enriched specificity group, indicating that pathogen cross-reactivity may be a feature of multiple cancers.

Tumor mutation burden is correlated with response and prognosis in microsatellite-stable (MSS) gastric cancer patients undergoing neoadjuvant chemotherapy

Summary: The combination of high tumor mutation burden (TMB-H) and 19q12 amplification (19q12 +) is associated with a favorable response to neoadjuvant chemotherapy (NACT) in microsatellite stable (MSS) gastric cancer patients. TMB-H is negatively correlated with lymph node metastasis and macrophage infiltration, and is a robust biomarker for better disease-free survival (DFS), which can be evaluated at the time of diagnosis. The findings were validated in two larger GC datasets, TCGA-STAD and ICGC-CN.
