The Multifaceted Regulation of Mitochondria in Ferroptosis

Summary: Ferroptosis is a novel form of regulated cell death initiated by lethal lipid peroxidation catalyzed by cellular labile free iron. It differs significantly from other regulated cell death forms at morphological, genetic, and biochemical levels. Dysregulated ferroptosis is highly relevant to the pathogenesis of degenerative diseases and organ injuries in human patients, and the targeted induction of ferroptosis is considered a potential therapeutic strategy for therapy-resistant cancers. Mitochondria play a crucial role in regulating ferroptosis, with compromised mitochondrial morphology and functionalities primarily involved in this process. Various mitochondria-associated proteins and metabolic processes have been identified to modulate ferroptotic cell death.
