Evolving Differentiation in African Trypanosomes

Juan F. Quintana et al.

Summary: Parasite differentiation is essential for adaptation to hosts and environments, with the transition between vertebrate and insect stages being well studied in African trypanosomes. Activation of advantageous metabolic pathways in the insect host leads to the formation of a 'stumpy' form. Various stimuli and stress conditions can trigger similar changes, suggesting a repurposing of a generic stress response for gains in parasite fitness during transmission.

Automated CUT&Tag profiling of chromatin heterogeneity in mixed-lineage leukemia

Derek H. Janssens et al.

Summary: This study maps fusion-specific targets of KMT2A oncofusion proteins across the genome, revealing aberrant chromatin regulation induced by these fusion proteins and cell-to-cell heterogeneity at these sites, which can predict sensitivity to therapeutic agents. Automated and single-cell CUT&Tag profiling characterizes the effects of KMT2A fusion proteins on chromatin in human primary leukemia samples, identifying oncogenic networks and fusion-specific therapeutic vulnerabilities.

Summary: In this study, we successfully adapted the CUT&Tag method to single-cell platforms for profiling chromatin landscapes at single-cell level. The analysis of H3K27me3 using scCUT&Tag distinguished cell types and allowed generation of cell-type-specific PcG landscapes.

A systematic analysis of Trypanosoma brucei chromatin factors identifies novel protein interaction networks associated with sites of transcription initiation and termination

Desislava P. Staneva et al.

Summary: Histones form nucleosomes around which DNA is wrapped to create chromatin, and histone post-translational modifications (PTMs) play a role in regulating gene expression. A study on Trypanosoma brucei identified proteins involved in histone acetylation and methylation, showing associations with specific regions of transcription initiation and termination. Proteomic analyses revealed novel components in the chromatin regulatory network of T. brucei, indicating divergence from conventional eukaryotic norms.

Single-cell transcriptomic analysis of bloodstream Trypanosoma brucei reconstructs cell cycle progression and developmental quorum sensing

Emma M. Briggs et al.

Summary: This study models the developmental steps of Trypanosoma brucei using oligopeptide-induced differentiation in vitro, capturing the transcriptomes of parasites through single cell transcriptomics. It details the relative order of biological events during asynchronous development, profiles dynamic gene expression patterns, identifies putative regulators, and provides a paradigm for dissecting differentiation events in parasites.

RSC primes the quiescent genome for hypertranscription upon cell-cycle re-entry

Christine E. Cucinotta et al.

Summary: This study found that there is robust transcription activity within the first few minutes of quiescence exit. The chromatin-remodeling enzyme RSC is already bound to genes induced upon quiescence exit. Depletion of RSC results in severe defects during quiescence exit, including a global decrease in Pol II loading and aberrant transcription initiation.
