A Causal-Pathway Phenotype of Chronic Fatigue Syndrome due to Hemodialysis in Patients with End-Stage Renal Disease

Halah Nori Asad et al.

Summary: This study aimed to investigate the associations between severity of fatigue and physio-somatic symptoms and glomerular filtration rate, inflammatory biomarkers, and Wnt/β-catenin pathway in ESRD patients. The results showed that ESRD patients exhibit high levels of fatigue and physio-somatic symptoms, which are associated with dialysis-induced changes in inflammatory pathways, the Wnt/β-catenin pathway, and copper levels.

Chronic fatigue syndrome and fibromyalgia-like symptoms are an integral component of the phenome of schizophrenia: neuro-immune and opioid system correlates

Rana Fadhil Mousa et al.

Summary: Physiosomatic symptoms play an important role in the phenomenology of schizophrenia. Higher scores of physiosomatic symptoms in schizophrenia patients are associated with increased levels of psychosis, hostility, excitement, formal thought disorders, psychomotor retardation, and negative symptoms. The variance in physiosomatic symptoms score is influenced by cognitive tasks, IL-10, IL-6, and other markers. Neural network analysis identified top predictors of physiosomatic symptoms including IL-6, HMGB1, education level, MOR, KOR, and IL-10. Immune pathways, neural markers, and alterations in the endogenous opioid system appear to contribute to the physiosomatic symptoms in schizophrenia.

Oxidative stress caused by a dysregulated Wnt/β-catenin signalling pathway is involved in abnormal placenta formation in pregnant mice with chronic fatigue syndrome

Hai Zhao et al.

Summary: Chronic fatigue syndrome (CFS) has a significant impact on embryonic development and placental morphology in pregnant mice, resulting in a decrease in the number of implantation sites for blastocysts, an increase in absorbed embryos, and abnormalities in placental structure. Additionally, CFS leads to increased oxidative stress, decreased antioxidase levels, and inhibition of the Wnt/beta-catenin signaling pathway in the placenta.

Study on the Correlation Between NF-κB and Central Fatigue

Xingzhe Yang et al.

Summary: Fatigue has been recognized as a major risk factor for human life and health, with nearly 1/3 of the population in Europe and America suffering from it. The NF-kappa B pathway plays a crucial role in central fatigue by regulating immune inflammatory responses and nerve functions. This review focuses on exploring the biological mechanisms of central fatigue and analyzing the clinical applications of NF-kappa B inhibitors in treating fatigue.

Inflammation and Wnt Signaling: Target for Immunomodulatory Therapy?

Imen Jridi et al.

Summary: Wnt proteins play crucial roles in development, cell fate specification, tissue regeneration, and tissue homeostasis, while aberrant Wnt signaling is associated with developmental defects, malignant transformation, inflammation, and cancer. Recent research suggests that dysregulated activation of Wnt signaling is involved in the pathogenesis of chronic inflammatory diseases, providing new insights for therapeutic interventions against inflammatory conditions.

Ashwagandha (Withania somnifera) root extract attenuates hepatic and cognitive deficits in thioacetamide-induced rat model of hepatic encephalopathy via induction of Nrf2/HO-1 and mitigation of NF-κB/MAPK signaling pathways

Heba M. A. Khalil et al.

Summary: The study demonstrated that Ashwagandha root extract improved symptoms of thioacetamide-induced hepatic encephalopathy, including liver toxicity indices, ammonia levels, and brain histopathological changes. Ashwagandha also reduced levels of oxidative stress markers and modulated the expression of key proteins and genes.

Wnt/β-Catenin Pathway in Experimental Model of Fibromyalgia: Role of Hidrox(R)

Ramona D'Amico et al.

Summary: The study investigated the potential beneficial effects of a new compound, Hidrox(R) (HD), in counteracting pain associated with fibromyalgia. Results showed that daily treatment with HD could modulate the WNT/beta-catenin and Nrf2 pathways to attenuate behavioral deficits and microglia activation induced by reserpine injection. This suggests that nutritional consumption of HD could be a new therapeutic approach for human fibromyalgia.

The Reification of the Clinical Diagnosis of Myalgic Encephalomyelitis/ Chronic Fatigue Syndrome (ME/CFS) as an Immune and Oxidative Stress Disorder: Construction of a Data-driven Nomothethic Network and Expo-sure of ME/CFS Subgroups

Michael Maes et al.

Summary: This study aimed to construct a model of ME/CFS using PLS path analysis and data on causome, protectome, and AOP. The results showed that 31.6% of the variance in physiosomatic symptoms of ME/CFS could be explained by various factors. Cluster analysis revealed three distinct immune groups within ME/CFS, each characterized by different immune responses.

Role of Kynurenine Pathway in Oxidative Stress during Neurodegenerative Disorders

Adrian Mor et al.

Summary: Neurodegenerative disorders have a negative impact on patients' quality of life, with genetic background, oxidative stress, and mitochondrial damage being partly responsible for their development. Activation of the kynurenine pathway leads to the accumulation of neuroactive and pro-oxidative metabolites, increasing oxidative stress and disrupting cellular energy metabolism. Modulating KP activity may help alleviate symptoms of neurodegenerative disorders.
