RGS12 Drives Macrophage Activation and Osteoclastogenesis in Periodontitis

Summary: Ablation of RGS12 in hematopoietic cells was shown to prevent bone loss in a periodontitis model, inhibit osteoclast formation and activity, and reduce inflammatory cytokine production in periodontal tissue. In vitro experiments also demonstrated that overexpression of RGS12 promoted proinflammatory macrophage reprogramming and enhanced migration ability, while knockdown of RGS12 inhibited inflammatory cytokine production and macrophage migration in response to stimuli like lipopolysaccharide. The results suggest that targeting RGS12 in macrophages could be a promising therapeutic approach for periodontitis.

Axin2+PDL Cells Directly Contribute to New Alveolar Bone Formation in Response to Orthodontic Tension Force

Summary: The Wnt-beta-catenin signaling pathway is crucial in orthodontic tooth movement, with Axin2(+) PDL cells being highly sensitive to orthodontic tension force and playing a critical role in PDL expansion and alveolar bone formation during the process. Targeting Axin2(+) PDL progenitor cells in future drug development may accelerate alveolar bone formation in orthodontic treatment.

Macrophage RGS12 contributes to osteoarthritis pathogenesis through enhancing the ubiquitination

Summary: This study identifies the role of ubiquitination in osteoarthritis (OA) and reveals the mechanism involving the protein RGS12 in promoting inflammation. The findings demonstrate that RGS12 enhances the association between ubiquitin and IKB, leading to the degradation of IKB through ubiquitination. Additionally, the increased ubiquitination inhibits the expression of MTAP, indirectly activating the phosphorylation of IKB. These processes ultimately result in the translocation of NF-KB to the nucleus and the upregulation of cytokine gene expression during inflammation.

Summary: The study revealed that regulation of regulator of G protein signaling 16 (RGS16) and G protein signaling in osteoblasts plays a crucial role in modulating the response of osteoblastic ovarian cancer G protein-coupled receptor 1 (OGR1) to metabolic acidosis and subsequent stimulation of osteoclastic bone resorption. Further investigations into the regulation of OGR1 in metabolic acidosis-induced bone resorption will aid in understanding bone loss in acidotic patients with chronic kidney disease.
