Unsuspected somatic mosaicism for FBN1 gene contributes to Marfan syndrome

Summary: This study reports on the experience of identifying five cases of mosaic pathogenic variants in the FBN1 gene among over 5,000 Marfan syndrome probands using next-generation sequencing technology. The rare findings of symptomatic MFS probands with a mosaic FBN1 pathogenic variant highlight the importance of thorough clinical examinations and regular follow-up for apparently asymptomatic mosaic parents. It is recommended to use NGS capture panels with adapted variant calling analyses for individuals with typical MFS who do not have identified single-nucleotide pathogenic variants or exon deletions/duplications.
