Summary: Ex vivo normothermic machine perfusion (NMP) of donor kidneys provides a platform for direct delivery of cellular therapeutics, such as Multipotent Adult Progenitor Cells (MAPC(R)), to optimize organ quality prior to transplantation. This study demonstrated improved urine output, decreased injury biomarker expression, and regulation of inflammatory factors in MAPC-treated kidneys during NMP, suggesting a promising strategy for organ reconditioning before transplantation.

Transient mTOR inhibition rescues 4-1BB CAR-Tregs from tonic signal-induced dysfunction

Summary: The study demonstrates that 4-1BB CAR-Tregs show more dramatic effects in activation and proliferation than CD28 CAR-Tregs, but exhibit reduced in vivo suppressive capacities. Transient exposure to a Treg stabilizing cocktail can sharply improve the in vivo function and expansion of 4-1BB CAR-Tregs. These findings suggest that transient mTOR inhibition can mitigate the negative effects of 4-1BB tonic signaling in Tregs.
