Identification of Functional HLA-A*01:01-Restricted Epstein-Barr Latent Membrane Protein 2-Specific T-Cell Receptors

Wesley Huisman et al.

Summary: In this study, we isolated the first functional HLA-A*01:01-restricted EBV-LMP2-specific T-cell populations and TCRs and demonstrated that these T-cell populations and T cells modified to express the LMP2-specific T-cell receptor showed IFN-gamma secretion and cytotoxicity toward EBV-LMP2-expressing malignant cell lines.

TCR Recognition of Peptide-MHC-I: Rule Makers and Breakers

Christopher Szeto et al.

Summary: T cells play a critical role in the adaptive immune system by distinguishing between healthy and unhealthy cells and aiding in clearing infections. The interaction between T cell receptors and peptide-MHC complexes is crucial for T cell activation, with recent structural biology studies providing new insights into this key event.

Magnitude of Off-Target Allo-HLA Reactivity by Third-Party Donor-Derived Virus-Specific T Cells Is Dictated by HLA-Restriction

Wesley Huisman et al.

Summary: Virus-specific CD8(pos) T cells derived from third-party donors exhibit high frequency and diversity of allo-HLA cross-reactivity, especially in HLA-B*08:01-restricted T cells. Thymic selection for HLA-B alleles significantly influences the level of allo-HLA cross-reactivity mediated by HLA-B*08:01-restricted T cells. These results suggest that the degree and specificity of allo-HLA cross-reactivity by T cells follow certain rules.
