Multiscale Simulation of an Influenza A M2 Channel Mutant Reveals Key Features of Its Markedly Different Proton Transport Behavior

Laura C. Watkins et al.

Summary: The D44N mutation in the influenza A M2 channel opens the channel gate and increases proton conduction, leading to a loss of asymmetric conduction. This mutation lowers the energy barrier for His37 deprotonation in the activated state and facilitates proton flux through the Va127 tetrad. Increased hydration around the His37 tetrad enhances proton transport efficiency.

Transportin-3 Facilitates Uncoating of Influenza A Virus

Jiahui Zou et al.

Summary: Influenza A viruses (IAVs) pose a significant global health threat, and this study explores the role of transportin-3 (TNPO3) in IAV infection. TNPO3 deficiency inhibits IAV replication by restraining the uncoating process and can interact with viral proteins M1 and M2.

Hydrophobic Residues at the Intracellular Domain of the M2 Protein Play an Important Role in Budding and Membrane Integrity of Influenza Virus

Danqi Bao et al.

Summary: The 91-94 hydrophobic motif of the M2 protein is crucial for the budding, bilayer membrane integrity, and pathogenicity of influenza viruses. Mutations that reduce the hydrophobicity of this motif significantly affect the budding ability of the M2 protein and impair the integrity of the viral membrane.

Influenza A virus protein PB1-F2 impairs innate immunity by inducing mitophagy

Wang Ruifang et al.

Summary: IAV infection induces mitophagy, with the PB1-F2 protein playing a key role in this process by translocating to mitochondria and interacting with TUFM and LC3B. PB1-F2-induced mitophagy leads to degradation of MAVS and suppression of type I interferon production, resulting in impaired cellular innate immunity, making it a potential therapeutic target.

ORF3a of the COVID-19 virus SARS-CoV-2 blocks HOPS complex-mediated assembly of the SNARE complex required for autolysosome formation

Guangyan Miao et al.

Summary: ORF3a of the COVID-19 virus SARS-CoV-2 inhibits autophagy activity by blocking fusion of autophagosomes/amphisomes with lysosomes. This leads to accumulation of autophagosomes/amphisomes, impaired lysosome function, and ultimately blocks autophagy process.

The immunodominant and neutralization linear epitopes for SARS-CoV-2

Shuai Lu et al.

Summary: The study reveals that predicted epitopes on SARS-CoV-2 effectively induce antibody production, with some being immunodominant. There are differences in immunodominant epitopes between individuals with domestic and imported SARS-CoV-2, possibly due to mutations on the proteins. Several epitopes on the S protein elicit neutralizing antibodies against different variants of SARS-CoV-2, showing potential for vaccine design against coronaviruses.

Foot-and-Mouth Disease Virus Inhibits RIP2 Protein Expression to Promote Viral Replication

Huisheng Liu et al.

Summary: The study found that RIP2 inhibited the replication of foot-and-mouth disease virus (FMDV) and played an important role in the activation of the IFN-β and NF-κB signal pathways during FMDV infection. Specific regions of FMDV proteins were responsible for reducing RIP2 protein expression, with specific regions of 2B and 2C being crucial for this process.

A viroporin-like 2B protein of duck hepatitis A virus 1 that induces incomplete autophagy in DEF cells

Zezheng Liu et al.

Summary: The study found that the 2B protein of Duck hepatitis A virus 1 can induce autophagy in host cells, but the autophagic flow is incomplete. This suggests that the infection and pathogenic mechanism of DHAV-1 may be related to autophagy.

Bluetongue virus non-structural protein 3 (NS3) and NS4 coordinatively antagonize type I interferon signaling by targeting STAT1

Zhuoran Li et al.

Summary: NS3 and NS4 of BTV coordinate to counteract IFN-I signaling in the JAK-STAT pathway by directly interacting with STAT1, targeting the SH2 domain of STAT1 to inhibit its phosphorylation. This study confirms the participation of NS3 and NS4 in interfering with IFN-I signaling and reveals a new mechanism used by BTV to evade host innate immune responses.

HIV-1 Vpu Promotes Phagocytosis of Infected CD4+ T Cells by Macrophages through Downregulation of CD47

Lijun Cong et al.

Summary: The HIV-1 accessory protein Vpu modulates macrophage infection by downregulating CD47 expression on infected cells, promoting phagocytosis. This mechanism enhances the susceptibility of macrophages to viral infection and allows for efficient infection by weakly macrophage-tropic virus strains. The findings provide new insights into intercellular transmission of HIV-1 to macrophages and have implications for viral reservoir establishment and early dissemination in vivo.

MARCH8 inhibits influenza A virus infection by targeting viral M2 protein for ubiquitination-dependent degradation in lysosomes

Xiaoman Liu et al.

Summary: MARCH8 acts as an important player in the host anti-influenza virus defense by targeting M2 protein and inhibiting viral replication. The discovery that H1N1 influenza A virus has evolved to resist MARCH8-mediated ubiquitination and degradation sheds light on the inhibitory effects of MARCH8 on virus transmission within the human population.
