Multifunctional intracellular matrix metalloproteinases: implications in disease

Wesam Bassiouni et al.

Summary: Matrix metalloproteinases (MMPs) are a family of zinc-dependent endopeptidases that were initially discovered as proteases targeting extracellular proteins. However, recent research has revealed their crucial intracellular roles in various pathological conditions, including cardiovascular renal disorders, inflammation, and malignancy. In addition to MMP-2, at least eleven other MMPs have been identified to function intracellularly, highlighting the importance of understanding their roles in cellular processes and potential as therapeutic targets.

Serum matrix metalloproteinase 7 (MMP7) is a biomarker of fibrosis in patients with non-alcoholic fatty liver disease

Katharine M. Irvine et al.

Summary: NAFLD affects 25% of the adult population globally. MMP7 has been identified as a serum biomarker of histological advanced fibrosis in NAFLD patients, improving diagnostic performance. Combining serum biomarkers has the potential to enhance sensitivity and specificity in detecting fibrosis in at-risk NAFLD patients.

Autoantibodies to speckled protein family in primary biliary cholangitis

Alessandro Granito et al.

Summary: The autoantibody profile of primary biliary cholangitis (PBC) includes antinuclear antibodies (ANA) which are detectable by indirect immunofluorescence in over 50% of PBC patients. One of the historically considered PBC-specific immunofluorescence patterns is the multiple nuclear dots (MND), targeting nuclear body proteins and suggesting potential novel therapeutic approaches for immune disorders like PBC.

Summary: The study identified a high specificity of anti-PML NB antibodies in diagnosing PBC. Positive reactivity of these antibodies was associated with higher concentrations of bilirubin and alkaline phosphatase. However, serum bilirubin remains the main prognostic marker for survival.

Summary: The development of a 'PBC Integrated Patient Care Pathway' by expert consensus simplifies and standardizes the management of PBC for clinicians based on current practice. This tool complements the EASL Clinical Practice Guidelines for the diagnosis and management of PBC patients, providing a practical decision tree and supporting the adoption of varied care provision models for patients with PBC.

Production and activity of matrix metalloproteinases during liver fibrosis progression of chronic hepatitis C patients

Moises Martinez-Castillo et al.

Summary: This study found that chronic hepatitis C patients had higher serum concentrations of MMPs compared to healthy individuals, with MMP-7 being able to distinguish early and advanced fibrosis stages. Collagenolytic activity was detected at F0 and F1, while gelatinase activity was not detected at any fibrosis stage. Tissue inhibitor of metalloproteinase-1 showed upregulation in F0 and F1 but downregulation in F2.
