Different Responses to Neoadjuvant Chemotherapy in Urothelial Carcinoma Molecular Subtypes

Gottfrid Sjodahl et al.

Summary: This study investigates the impact of molecular subtypes on pathological response and survival in patients with muscle-invasive bladder cancer (MIBC) receiving preoperative cisplatin-based chemotherapy. The results suggest that patients with certain molecular subtypes, such as genomically unstable (GU) and urothelial-like (Uro) tumors, are more responsive to cisplatin-based neoadjuvant chemotherapy and have improved survival compared to those with the basal/squamous (Ba/Sq) subtype. The study highlights the potential of subtype-specific biomarkers, such as osteopontin (SPP1), for developing a more precision-based treatment approach for neoadjuvant chemotherapy in MIBC.

Identification of the pyroptosis-related prognostic gene signature and the associated regulation axis in lung adenocarcinoma

Wanli Lin et al.

Summary: A prognostic PRG signature containing five genes (NLRP7, NLRP1, NLRP2, NOD1, and CASP6) was identified for LUAD patients through comprehensive bioinformatics analysis. Additionally, a lncRNA KCNQ1OT1/miR-335-5p/NLRP1/NLRP7 regulatory axis was discovered, which may play a significant role in the progression of LUAD. Further research is needed to validate these findings.

Rebecca L. Siegel et al.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Channelling inflammation: gasdermins in physiology and disease

Xing Liu et al.

Summary: Gasdermins have been identified as mediators of inflammatory cell death known as pyroptosis, forming cell membrane pores upon activation to release pro-inflammatory cytokines. They play important roles in autoimmune and inflammatory diseases, infectious diseases, deafness, and cancer, with potential for novel therapeutic avenues.

Galectin-9 interacts with PD-1 and TIM-3 to regulate T cell death and is a target for cancer immunotherapy

Riyao Yang et al.

Summary: The study reveals that PD-1 contributes to exhausted T cell survival by binding to TIM-3 ligand Gal-9 and inhibiting Gal-9/TIM-3 induced cell death. Anti-Gal-9 therapy selectively expands intratumoral TIM-3(+) cytotoxic CD8 T cells and T-reg cells, while combination treatment with anti-Gal-9 and GITR agonist shows synergistic antitumor activity.

A novel defined pyroptosis-related gene signature for predicting the prognosis of ovarian cancer

Ying Ye et al.

Summary: This study identified pyroptosis-related genes in ovarian cancer (OC) that may play important roles in tumor immunity and can be used to predict the prognosis of OCs. By dividing OC cases into subtypes based on the expression of these genes, different risk groups were identified with significant differences in survival possibilities. Additionally, immune-related genes were found to be enriched in high-risk groups, indicating a decreased immune status in these patients.

Fibroblast growth factor receptor 3 alterations and response to immune checkpoint inhibition in metastatic urothelial cancer: a real world experience

Tracy L. Rose et al.

Summary: Our study confirms that both FGFR3-altered and wild type bladder cancers show equal response to immune checkpoint blockade (ICB). Despite the lower inflammation in FGFR3-altered tumors, they exhibit similar T cell receptor diversity as non-altered tumors. The balance of CD8 T cell gene expression signature to immune suppressive features plays a crucial role in determining the response to ICB.

Integrative analysis of prognostic long non-coding RNAs with copy number variation in bladder cancer

Wenwen Zhong et al.

Summary: This study identified prognosis-related lncRNAs in bladder cancer by analyzing the expression profiles of lncRNAs and CNVs, and revealed a risk-score signature associated with overall survival in BLCA through multi-omics integration analysis. The enrichment analysis indicated that the imbalance in MAPK signaling pathway, focal adhesion pathway, and JAK-STAT signaling pathway is closely related to tumor development in BLCA.

Pyroptosis in Cancer: Friend or Foe?

Xiuxia Lu et al.

Summary: Pyroptosis, a new form of programmed cell death, has garnered attention in cancer research. Various stimuli can trigger pyroptosis, but its role in cancer remains unclear with conflicting findings. Despite the uncertainties, pyroptosis may hold potential as a promising anti-cancer therapy.

The Pyroptosis-Related Signature Predicts Prognosis and Indicates Immune Microenvironment Infiltration in Gastric Cancer

Summary: This study classified gastric cancer patients based on pyroptosis-related regulators and developed a prognostic model called PS-score, which can predict patients' prognosis and overall survival time. Patients with a low PS-score have more inflammatory cell infiltration and better response to PD1/PD-L1 blockade immunotherapy.
