Long noncoding RNA ADIRF antisense RNA 1 upregulates insulin receptor substrate 1 to decrease the aggressiveness of osteosarcoma by sponging microRNA-761

Lingling Xu et al.

Summary: ADIRF-AS1 is overexpressed in osteosarcoma (OS) and acts as a competitive endogenous RNA, sequestering miR-761 and leading to overexpression of its target gene IRS1. Knockdown of ADIRF-AS1 inhibits proliferation, migration, and invasiveness of OS cells and promotes apoptosis. These findings may contribute to the development of lncRNA-directed therapeutics for OS.

Cracking the Breast Cancer Glyco-Code through Glycan-Lectin Interactions: Targeting Immunosuppressive Macrophages

Nuno Lopes et al.

Summary: The immune microenvironment of breast cancer is influenced by tumor-associated macrophages (TAM), which play a critical role in immune suppression and cancer progression. Recent studies have identified altered glycan structures on breast cancer cells as potential targets for novel immunotherapies, highlighting the potential of targeting glycan-lectin interactions to reshape the immunosuppressive landscape of breast cancer.

Actin-Like Protein 8 Promotes the Progression of Triple-Negative Breast Cancer via Activating PI3K/AKT/mTOR Pathway

Shaoxia Fan et al.

Summary: In this study, it was found that ACTL8 is upregulated in TNBC and is associated with poor prognosis. Silencing ACTL8 inhibited proliferation, migration, and invasion while promoting apoptosis in MDA-MB-231 and BT-549 cells. Additionally, silencing ACTL8 suppressed the activation of the PI3K/AKT/mTOR signaling pathway in these cells, indicating a potential mechanism of ACTL8 in TNBC progression.

Optimal anchoring of a foldamer inhibitor of ASF1 histone chaperone through backbone plasticity

Johanne Mbianda et al.

Summary: The study demonstrates that the structural plasticity of foldamer backbone is crucial for optimal interaction with protein surface. By designing a foldamer/peptide hybrid inhibitor using oligoureas as α-helix mimics, effective inhibition of the key regulator of chromatin dynamics, histone chaperone ASF1, was achieved. Additionally, the ASF1 ligands with nonpeptide oligourea segments showed significantly improved resistance to proteolysis in human plasma compared to alpha-helical peptides.

Comprehensive analysis of pan-cancer reveals potential of ASF1B as a prognostic and immunological biomarker

Xinyao Hu et al.

Summary: The study showed that ASF1B is highly expressed in 22 cancers and is negatively correlated with the prognosis of multiple major cancer types. ASF1B expression was also correlated with TMB in 21 cancers and with MSI in 7 cancers. ASF1B may potentially serve as a prognostic marker and potential immunotherapeutic target for several malignancies due to its role in tumorigenesis and immune infiltration.

CircUBAP2 Promotes MMP9-Mediated Oncogenic Effect via Sponging miR-194-3p in Hepatocellular Carcinoma

Boqiang Liu et al.

Summary: This study identified a novel circRNA, circUBAP2, in hepatocellular carcinoma (HCC) through bioinformatics analysis. CircUBAP2 was significantly upregulated in HCC tissues and correlated with aggressive clinical characteristics and poor prognosis. Functionally, circUBAP2 promoted cell proliferation, colony formation, migration, and invasion in vitro, and enhanced tumor growth and pulmonary metastasis in vivo. Mechanistically, circUBAP2 acted as a competing endogenous RNA (ceRNA) for miR-194-3p, thereby regulating the expression of MMP9 and promoting HCC progression. CircUBAP2 could potentially serve as a prognostic biomarker and therapeutic target for HCC patients.
