Microfluidic production and characterization of biofunctionalized giant unilamellar vesicles for targeted intracellular cargo delivery

Summary: Researchers have developed a microfluidic-based mechanical droplet-splitting pipeline for the production of carrier-GUVs with diameters of around 2 μm, allowing for efficient cargo loading and precise control over the properties of GUV membranes. By investigating GUV-cell interactions, they achieved targeted cellular GUV delivery and explored strategies for intracellular GUV cargo release through lysosomal escape. This delivery technology could lead to more efficient drug administration and enable targeted delivery of advanced cargo like microparticles, viruses, or macromolecular DNA-robots.

Summary: ExoIL12, a novel engineered exosome therapeutic displaying functional IL12 on its surface, shows prolonged tumor retention, enhanced antitumor activity, and increased tumor antigen-specific CD8(+) T cells compared to rIL12. With 10-fold higher intratumoral exposure and prolonged IFN gamma production, ExoIL12 outperforms rIL12 in tumor growth inhibition and immune memory, providing a therapeutic window for this potent cytokine.

Summary: Extracellular vesicles play a crucial role in intercellular communication and cell physiology, but quantitative knowledge on their signaling mechanisms is lacking, hampering their therapeutic applications. By using a synthetic approach, researchers have shown that synthetic EVs have functionalities similar to natural EVs and can be used for therapeutic purposes in wound healing and neovascularization. Transcriptome profiling has enabled a systematic decoding of synergistic effects between individual EV constituents, leading to a better understanding of EV signaling.
