Facile and label-free fluorescence strategy for evaluating the influence of bioactive ingredients on FMO3 activity via supramolecular host-guest reporter pair

Summary: A facile and label-free fluorescence strategy was developed to evaluate the influence of bioactive compounds in Xuefu Zhuyu decoction (XFZYD) on flavin monooxygenase 3 (FMO3) activity, revealing potential inhibitors of FMO3 from Chinese traditional medicine. Naringin, paeoniflorin, beta-ecdysterone, 18 beta glycyrrhizic acid, amygdalin, albiflorin, and saikosaponin A were identified as compounds that downregulated FMO3 activity and reduced TMAO biosynthesis, with molecular docking used to simulate the receptor-ligand complex conformation. This study presents a novel and promising approach for screening FMO3 inhibitors from Chinese traditional medicine through supramolecular sensing.

Baicalein alleviates hyperuricemia by promoting uric acid excretion and inhibiting xanthine oxidase

Summary: Baicalein (BAL) has potent anti-hyperuricemic effects by promoting renal urate excretion and inhibiting serum uric acid production through downregulating GLUT9 and URAT1 expression as well as suppressing XOD activity.
