Rapid and High-Throughput Evaluation of Diverse Configurations of Engineered Lysins Using the VersaTile Technique

Summary: Bacteriophage-encoded lysins are a novel class of antibacterial enzymes that degrade peptidoglycan. The modular composition of lysins allows for optimization of antibacterial properties, with new techniques such as VersaTile facilitating the rapid construction of large lysin libraries. Through high-throughput screening of combinatorial lysin libraries targeting Klebsiella pneumoniae, specific design rules were identified, with outer membrane permeabilizing peptides (OMPs) and enzymatically active domains (EADs) being overrepresented among the top hits.
