Structure Guided Design of Bacteriophage Qβ Mutants as Next Generation Carriers for Conjugate Vaccines

Summary: Vaccines are critical tools for treating and preventing diseases. However, there are few available carriers for effective conjugate vaccines, and current protein carriers often induce high levels of antibodies against themselves, interfering with immune responses. Researchers have obtained the crystal structure of a promising protein carrier called bacteriophage Qfl virus-like particle (mQfl), and designed mutants of mQfl that can elicit strong immune responses against tumor antigens while producing lower levels of anti-carrier antibodies. In mouse models, immunization with mQfl-TACA conjugates resulted in a significant reduction in tumor growth compared to unimmunized mice. Moreover, mQfl has shown potential as a carrier for developing vaccines against COVID-19, inducing high levels of IgG antibodies against peptide antigens from the SARS-CoV-2 virus. Structure-based rational design is a powerful strategy for developing new vaccine carriers.
