Opportunistic use of catecholamine neurotransmitters as siderophores to access iron by Pseudomonas aeruginosa

Quentin Perraud et al.

Summary: Iron is a crucial nutrient for bacterial growth and plays a key role in the battle between pathogens and hosts during infection. Pseudomonas aeruginosa utilizes siderophores to access iron from the host, and catecholamine neurotransmitters are shown to effectively transport iron into bacterial cells. Specific transporters PiuA and PirA play pivotal roles in this process, with different preferences for catecholamines uptake.

68Ga-labelled desferrioxamine-B for bacterial infection imaging

Milos Petrik et al.

Summary: In this study, DFO-B was successfully radiolabeled with (68)Ga and showed promising potential for specific imaging of bacterial infections both in vitro and in vivo, with high specificity and minimal off-target effects. Considering its long clinical use and low estimated radiation dose, (68)Ga-DFO-B could be a valuable tool for clinical translation in the future.

Vitamin B12-peptide nucleic acids use the BtuB receptor to pass through the Escherichia coli outer membrane

Tomasz Pienko et al.

Summary: By covalently linking vitamin B-12 with PNA, researchers were able to utilize the vitamin B-12 transport system to deliver PNA to E. coli cells. The conjugate follows the same route through the outer membrane as free vitamin B-12, with BtuB playing a crucial role in the transport process. Molecular dynamics simulations provided insights into the mechanism of conjugate permeation through BtuB, highlighting the potential for hijacking the vitamin B-12 system for efficient PNA delivery.

Harnessing Iron Acquisition Machinery to Target Enterobacteriaceae

Artur Sargun et al.

Summary: The review outlines the use of siderophore-mediated iron acquisition in Gram-negative bacteria, providing a foundation for the conceptualization and development of approaches to prevent and treat bacterial infections. Differences in the siderophore-based iron uptake mechanisms between pathogenic Enterobacteriaceae and commensal microbes may lead to the development of selective Trojan-horse antimicrobials and immunization strategies that will not harm the host microbiota.

Antibiotic Conjugates with an Artificial MECAM-Based Siderophore Are Potent Agents against Gram-Positive and Gram-Negative Bacterial Pathogens

Lukas Pinkert et al.

Summary: The urgent medical need for novel drugs against Gram-negative bacteria led to the synthesis of antibiotics and artificial siderophores conjugates, with predicted structures and iron binding properties in silico. MECAM demonstrated effective artificial siderophore activity in Escherichia coli, and after linker optimization, certain conjugates showed potent growth inhibition of multidrug-resistant pathogens. The uptake pathway of MECAMs was deciphered, with resistance against one conjugate mediated by a mutation in the gene encoding ExbB involved in siderophore transport.

The Esterase PfeE, the Achilles' Heel in the Battle for Iron between Pseudomonas aeruginosa and Escherichia coli

Veronique Gasser et al.

Summary: Bacteria access iron through siderophores, but Pseudomonas aeruginosa can also pirate siderophores produced by other microorganisms. Deletion of the pfeE gene in P. aeruginosa prevents it from accessing iron through enterobactin. Under iron-restricted conditions, P. aeruginosa strongly represses E. coli growth as long as it can produce its own siderophores, but if it cannot, it relies on enterobactin produced by E. coli. If pfeE is deleted, E. coli gains the upper hand in the culture.

Cefiderocol, a new antibiotic against multidrug-resistant Gram-negative bacteria

Jose Tiago Silva et al.

Summary: Cefiderocol is a novel catechol-substituted siderophore cephalosporin that overcomes resistance mechanisms of Gram-negative bacteria by binding to extracellular free iron and using bacterial active iron transport channels. Despite higher all-cause mortality observed in the CREDIBLE-CR trial among patients receiving Cefiderocol treatment, especially those with severe infections due to Acinetobacter spp, further phase III clinical studies are needed to evaluate its efficacy in the treatment of serious infections.

Efficient chito-oligosaccharide utilization requires two TonB-dependent transporters and one hexosaminidase in Cellvibrio japonicus

Estela C. Monge et al.

Summary: This study focused on the mechanisms of chitin utilization by the bacterium Cellvibrio japonicus, emphasizing the importance of chito-oligosaccharides in its metabolism. By combining transcriptomics and bacterial genetics, essential mechanisms for chito-oligosaccharide transport and catabolism in C. japonicus were determined, providing insights into polysaccharide degradation enzymes and transporters essential for chito-oligosaccharide utilization. This research further advances our understanding of C. japonicus chitin metabolism and may have implications for other environmentally or industrially important bacteria.

Yahiya Y. Syed

Summary: Cefiderocol is a cephalosporin with activity against carbapenem-resistant bacteria and nonfermenters, using the bacteria's iron uptake system to disrupt the cell wall and kill the bacteria. It has shown good effectiveness and safety in clinical trials for various serious bacterial infections, providing a useful addition to treatment options for adults with limited choices.

Optimization of Artificial Siderophores as 68Ga-Complexed PET Tracers for In Vivo Imaging of Bacterial Infections

Carsten Peukert et al.

Summary: The diagnosis of bacterial infections at deep body sites can benefit from noninvasive imaging techniques utilizing molecular probes labeled with iron transport-specific siderophores and PET technology. Probes containing gallium-68 linked to iron-binding siderophores were found to be suitable for in vivo imaging of bacteria, showing promise in discerning infection from inflammation.
