Nanocatalytic Theranostics with Glutathione Depletion and Enhanced Reactive Oxygen Species Generation for Efficient Cancer Therapy

Lian-Hua Fu et al.

Summary: Chemodynamic therapy (CDT) is a promising method for killing cancer cells by converting intracellular hydrogen peroxide (H2O2) into highly toxic hydroxyl radicals ((OH)-O-center dot). An intelligent nanocatalytic theranostics, PGC-DOX, was developed to address the limitations of endogenous H2O2 and high GSH levels in tumor cells, leading to efficient cancer therapy. The system combines H2O2 self-supply, GSH-elimination, and DOX-induced chemotherapy to effectively inhibit tumor growth with minimal side effects in vivo.

Doxorubicin-loaded hydrogen peroxide self-providing copper nanodots for combination of chemotherapy and acid-induced chemodynamic therapy against breast cancer

Jie-xia Li et al.

Summary: The study introduced H2O2 self-providing copper nanodots and successfully developed DOX-nanodots, showing promising therapeutic effects against breast cancer with enhanced cell apoptosis and decreased cell proliferation. The treatment exhibited good biocompatibility with no obvious toxicity, potentially due to the specific OH generation in the weakly acidic tumor microenvironment. This approach holds potential in cancer treatment due to its tumor-specific production of reactive oxygen species and successful combination with chemotherapy.

A Ferrocene-Functionalized Covalent Organic Framework for Enhancing Chemodynamic Therapy via Redox Dyshomeostasis

Le-Le Zhou et al.

Summary: A nanodrug, RSL3@COF-Fc (2b), based on covalent organic framework (COF) has been fabricated to enhance cellular damage induced by CDT through promoting Fenton-like reactions and inhibiting GPX4 irreversibly. This approach synergistically results in massive lipid peroxide accumulation, subsequent cell damage, and ultimately ferroptosis, not being limited by intracellular glutathione, providing new insights for developing COF-based nanomedicine.
