A clinically applicable and scalable method to regenerate T-cells from iPSCs for off-the-shelf T-cell immunotherapy

Shoichi Iriguchi et al.

Summary: The study presents a feeder-free, scalable differentiation process of human induced pluripotent cells (iPSCs) into T-cells with T-cell receptor dependent anti-tumor function in vitro and in vivo, providing a platform for clinical application and research on human T-cell differentiation and biology.

DL4-μbeads induce T cell lineage differentiation from stem cells in a stromal cell-free system

Ashton C. Trotman-Grant et al.

Summary: A stromal cell-free, microbead-based approach has been developed to efficiently generate T-lineage cells from various cell sources, including cord blood, GCSF-mobilized peripheral blood, and pluripotent stem cells. The method involves the use of DL4-mu beads and lymphopoietic cytokines to induce a sequential differentiation process leading to the development of functional T cells. Single-cell RNA sequencing of human pluripotent stem cell-derived proT cells shows a transcriptional profile similar to early thymocytes. Adoptive transfer of CD34(+)CD7(+) proT cells into immunodeficient mice demonstrates successful thymic engraftment and maturation of peripheral T cells. The DL4-mu bead platform provides a simple and robust tool for studying human T cell development and developing engineered T cell therapies from sustainable sources.

Generation of macrophages with altered viral sensitivity from genome-edited rhesus macaque iPSCs to model human disease

Yoshihiro Iwamoto et al.

Summary: Researchers successfully established iPSC from non-human primates, differentiated them into functional macrophages, and knocked out TRIM5 gene in Rh-iPSCs using CRISPR-Cas9, showing an increased sensitivity to HIV infection in vitro. This protocol can be applied to expand NHP gene therapy models.
