Stress decreases serotonin tone in the nucleus accumbens in male mice to promote aversion and potentiate cocaine preference via decreased stimulation of 5-HT1B receptors

Harrison M. Fontaine et al.

Summary: The stress-induced release of dynorphins activates kappa opioid receptors in serotonergic neurons, leading to dysphoria and potentiation of drug reward. Anatomically and functionally distinct populations of KOR-expressing neurons in the dorsal raphe nucleus were identified. Optogenetic manipulation of these neurons modulated cocaine conditioned place preference, while excision of Pdyn from the medial nucleus accumbens blocked stress-potentiation of cocaine CPP. Activation of 5-HT1B receptors within the mNAc by Dyn/KOR is implicated in regulating stress response, affect, and drug reward.

The mixed kappa and delta opioid receptor agonist, MP1104, attenuates chemotherapy-induced neuropathic pain

Diana Vivian Atigari et al.

Summary: Addressing the urgent need for effective treatments for chronic pain, research suggests that mixed opioid receptor agonists targeting KOP and DOP could provide non-addictive pain medications with a wider therapeutic index. A novel compound, MP1104, demonstrates potent antinociceptive effects mediated via KOP and DOP without inducing rewarding or aversive effects.

Experience-dependent myelination following stress is mediated by the neuropeptide dynorphin

Lindsay A. Osso et al.

Summary: This study reveals that the neuropeptide dynorphin, released upon high levels of activity, promotes myelination. Following forced swim stress, an increase in OPC differentiation and myelination in related neurons is observed.

Kappa opioid receptor controls neural stem cell differentiation via a miR-7a/Pax6 dependent pathway

Summary: This study reveals that OPRK1 agonists inhibit adult neurogenesis in the mouse hippocampus by upregulating the expression of miR-7a, which in turn downregulates Pax6/Neurog2/NeuroD1 activities and hinders neuronal differentiation of neural stem cells.
