Endothelial cellmetabolicmemory causes cardiovascular dysfunction in diabetes

Yufeng Yao et al.

Summary: This study aimed to identify the molecular mechanism for hyperglycemia-induced metabolic memory in endothelial cells (ECs) and to demonstrate its critical importance in the development of cardiovascular dysfunction in diabetes. The results showed that hyperglycemia leads to dysregulation of multiple signaling pathways in ECs, which are remembered by the cells even under low glucose conditions and contribute to the development of cardiovascular disease. The study also identified a novel signaling pathway involved in metabolic memory.

Cardioprotective mechanism of SGLT2 inhibitor against myocardial infarction is through reduction of autosis

Kai Jiang et al.

Summary: SGLT2 inhibitor Empagliflozin (EMPA) has been shown to suppress cardiomyocyte autophagic cell death, reduce infarct size, improve cardiac function and survival, by inhibiting Na+/H+ exchanger 1 (NHE1) activity and regulating excessive autophagy. These findings provide new insights for drug development targeting NHE1 and autophagy for ventricular remodeling and heart failure treatment after myocardial infarction in both diabetic and non-diabetic patients.

Cardiovascular effects of non-insulin glucose-lowering agents: a comprehensive review of trial evidence and potential cardioprotective mechanisms

Summary: Type 2 diabetes mellitus is highly prevalent and associated with increased mortality due to cardiovascular diseases. New glucose-lowering agents require cardiovascular outcome trials, with DPP4i showing safety but limited improvement in mortality/morbidity, while GLP1-Ra and SGLT2i have shown to reduce cardiovascular risk.

Epigenetic Mechanisms in Diabetic Vascular Complications and Metabolic Memory: The 2020 Edwin Bierman Award Lecture

Summary: Diabetes complications such as macrovascular and microvascular complications contribute to increased mortality rates. Inflammation, oxidative stress, and fibrosis are common features, and epigenetic changes play a significant role in impacting gene expression. Environmental factors and lifestyle choices, along with epigenetic modifications like DNA methylation, are key factors in influencing diabetic conditions.

Association of CX3CR1 Gene Polymorphisms with Fractalkine, Fractalkine Receptor, and C-Reactive Protein Levels in Patients with Kidney Failure

Lukasz Wozny et al.

Summary: Fractalkine is a chemokine that plays a significant role in inflammation and has been studied in relation to chronic kidney disease. This study found that CX3CR1 and CRP levels were higher in ESRD patients compared to controls, while fractalkine levels were significantly elevated in ESRD patients with specific genetic polymorphisms. These findings suggest that CX3CR1 gene polymorphisms may be involved in the pathogenesis of ESRD.

Impaired brain fractalkine-CX3CR1 signaling is implicated in cognitive dysfunction in diet-induced obese mice

Namiko Kawamura et al.

Summary: The study showed that DIO mice exhibited significant cognitive deficits in the Y-maze test and decrease in fractalkine and CX3CR1 in the hippocampus and amygdala compared with mice fed a control diet. Administration of the CX3CR1 antagonist 18a in normal mice induced significant cognitive deficits in the Y-maze test.

Regulation and function of CX3CR1 and its ligand CX3CL1 in kidney disease

Sibylle von Vietinghoff et al.

Summary: The chemokine CX3CL1 and its receptor CX3CR1 play crucial roles in kidney inflammation, facilitating leukocyte attraction to the kidney and showing high expression in various kidney diseases. This axis functions in both localized and systemic effects of renal inflammation, particularly in regulating leukocyte activity in inflammatory conditions.

SGLT-2 inhibitors or GLP-1 receptor agonists for adults with type 2 diabetes: a clinical practice guideline

Sheyu Li et al.

Summary: A panel of experts issued risk-stratified recommendations for the use of SGLT-2 inhibitors or GLP-1 receptor agonists in adults with type 2 diabetes, based on their cardiovascular and kidney disease risk factors. The recommendations highlighted the benefits and potential harms of these medications in different risk groups, with a strong recommendation for starting SGLT-2 inhibitors in patients with established CVD and CKD. The recommendation also emphasized the importance of considering individual patient values and preferences in treatment decisions.

Summary: SGLT2 inhibitors and GLP-1 receptor agonists are effective in lowering glucose, weight loss, and blood pressure reduction, offering additional protection for cardiovascular disease in patients with type 2 diabetes. They are considered second-line therapies for diabetes, with prioritized consideration based on individual patient characteristics.

Summary: The study investigated the anti-inflammatory effects of empagliflozin and gemigliptin in macrophages, with a focus on signaling pathways. Results showed that combined treatment with empagliflozin and gemigliptin exhibited stronger anti-inflammatory properties, indicating potential benefits in tackling metabolic syndromes in chronic inflammatory diseases.
