Pulsatilla decoction alleviates colitis by enhancing autophagy and regulating PI3K-Akt-mTORC1 signaling pathway

Xuewei Wang et al.

Summary: The present study aimed to investigate the therapeutic effect of Pulsatilla decoction (PD) on ulcerative colitis (UC) and to elucidate its potential molecular mechanisms. The results showed that PD could reduce disease activity index and infiltration of NK1.1 cells in colitis mice, decrease the secretion of IL-13, and alleviate tissue inflammation and autophagy through modulating the PI3K-Akt-mTORC1 signaling pathway.

Mast Cell Tryptase Promotes Inflammatory Bowel Disease-Induced Intestinal Fibrosis

Bin Liu et al.

Summary: In this study, it was found that mast cell tryptase promotes inflammatory bowel disease-induced intestinal fibrosis by activating the PAR-2/Akt/mTOR pathway of fibroblasts. Tryptase stored in mast cell granules is released into fibrotic intestinal tissues, leading to increased expression of tryptase and differentiation of fibroblasts into fibrotic-phenotype myofibroblasts. Inhibition of tryptase reduced intestinal fibrosis, indicating the potential for targeting tryptase as a therapeutic strategy for intestinal fibrosis.

Isolation and characterization of phellodendronoside A, a new isoquinoline alkaloid glycoside with anti-inflammatory activity from Phellodendron chinense Schneid

Yanpo Si et al.

Summary: Phellodendronoside A isolated from Phellodendron chinense demonstrates excellent anti-inflammatory activity by inhibiting the overproduction of pro-inflammatory mediators and suppressing the activation of MAPK pathways. This suggests that PDA may be a potential agent for the treatment of inflammatory illnesses.

Anemoside B4 ameliorates TNBS-induced colitis through S100A9/MAPK/NF-κB signaling pathway

Yong Zhang et al.

Summary: The study demonstrated that anemoside B4 ameliorated TNBS-induced colitis symptoms by inhibiting tissue damage, inflammatory cell infiltration, pro-inflammatory cytokine production, apoptosis, and slowed proliferation in colon. Quantitative proteomic analyses revealed that anemoside B4 significantly altered 56 proteins in TNBS-induced rats, with a focus on proteins related to TCA cycle and respiratory electron transport chain. S100A9, one of the most significantly down-regulated proteins, was associated with NF-kappa B and MAPK signaling pathways in UC pathogenesis.

NGS study of glucocorticoid response genes in inflammatory bowel disease patients

Marzena Skrzypczak-Zielinska et al.

Summary: This study aimed to identify genetic predictors of variable reactions to glucocorticoid therapy in inflammatory bowel disease patients. By analyzing 21 genes in 139 patients, NR3C1, FKBP5, MAPK14, and ABCB1 were identified as significant factors affecting treatment response. These findings may serve as valuable pharmacogenetic biomarkers pending confirmation in other populations.

Novel mechanisms and clinical trial endpoints in intestinal fibrosis*

Jie Wang et al.

Summary: The incidence of inflammatory bowel diseases poses a global public health challenge, with intestinal fibrosis being a common complication in Crohn's disease. Recent progress has implicated mesenchymal cells, cytokines, microbial products, and mesenteric adipocytes in the pathogenesis of intestinal fibrosis. This information suggests that innovative therapies validated by reliable clinical trial endpoints may prevent or reverse intestinal strictures.
