Histone lactylation: epigenetic mark of glycolytic switch

Xiaofeng Dai et al.

Summary: Y histone lactylation and acetylation compete for epigenetic modification and mark the levels of lactates and acetyl-CoA. The choice between lactate and acetyl-CoA generation from glycolysis determines cell fate towards malignancy. Controlling the glycolytic switch marked by lactylation may offer novel therapeutic opportunities against cancer.

Lung Myofibroblasts Promote Macrophage Profibrotic Activity through Lactate-induced Histone Lactylation

Huachun Cui et al.

Summary: This study demonstrates that in lung fibrosis, myofibroblasts enhance glycolysis to produce lactate, which regulates the pathogenic phenotype of alveolar macrophages. Lactate induces profibrotic gene expression in macrophages, promoting the fibrotic process.

Function and Mechanism of Novel Histone Posttranslational Modifications in Health and Disease

Huiwen Xu et al.

Summary: Histone posttranslational modifications (HPTMs) regulate gene expression by modulating chromatin states and play significant roles in the pathogenesis of diseases. Novel HPTMs such as propionylation, butyrylation expand the modification repertoire, regulating cellular processes like chromatin remodeling and cell cycle to control cell functions.

Lactylation, a Novel Metabolic Reprogramming Code: Current Status and Prospects

Summary: Lactate is a product of glycolysis and serves as a critical energy source for mitochondrial respiration. It also acts as a precursor of gluconeogenesis and a signaling molecule. Studies have shown that lactate-mediated reprogramming of immune cells and enhancement of cellular plasticity contribute to establishing disease-specific immunity status. Further research on the relationship between lactate and histone modification is crucial for understanding disease progression and drug resistance.
