MiR-206 suppresses the deterioration of intrahepatic cholangiocarcinoma and promotes sensitivity to chemotherapy by inhibiting interactions with stromal CAFs

Renjie Yang et al.

Summary: Intrahepatic cholangiocarcinoma (iCCA) is a highly malignant subtype of cholangiocarcinoma (CCA) with poor prognosis. This study found that miR-206 is downregulated in iCCA tissues and its overexpression can inhibit cell proliferation, migration and invasion. Normal fibroblast cells can transform into cancer-associated fibroblast cells (CAFs) and promote malignant behavior and drug resistance in cancer cells. Overexpressing miR-206 can inhibit this mutual promoting effect and suppress tumor deterioration through exosomes.

More than a Bubble: Extracellular Vesicle microRNAs in Head and Neck Squamous Cell Carcinoma

Wittaya Panvongsa et al.

Summary: MicroRNAs (miRNAs) are small non-coding RNA molecules that play a key regulatory role in various biological processes. Dysregulation of miRNAs is associated with human diseases, particularly cancer. Extracellular vesicles (EVs) are important for intercellular communication, and miRNAs carried by EVs are involved in cell-to-cell interactions, including tumor development. In recent years, there has been rapid growth in understanding the molecular mechanisms and clinical importance of EV-miRNAs in head and neck squamous cell carcinoma (HNSCC). EV-miRNAs have multiple effects on HNSCC tumorigenesis, including tumor proliferation, invasion and metastasis, immune modulation, and treatment resistance. EV-miRNAs also have the potential to be used as biomarkers for screening, early diagnosis, and disease monitoring in liquid biopsies. Furthermore, EV-miRNAs hold promise as a novel therapeutic target.

The Potential Roles of Exosomal Non-Coding RNAs in Hepatocellular Carcinoma

Wei Wang et al.

Summary: Hepatocellular carcinoma (HCC) is a highly prevalent and deadly cancer with low diagnostic rates. Exosomes, as extracellular vesicles released by various cells, including tumor cells, play a crucial role in HCC progression. Non-coding RNAs (ncRNAs) in exosomes have been identified as important regulators of tumor development. In this review, we discuss the formation and uptake of exosomes, classification of exosomal ncRNAs, and the current research on their roles in HCC progression. Additionally, we explore the potential clinical applications of exosomal ncRNAs as diagnostic biomarkers and therapeutic targets in HCC.

Exosomal microRNA-15a from mesenchymal stem cells impedes hepatocellular carcinoma progression via downregulation of SALL4

Yu-Shui Ma et al.

Summary: Exosomal miR-15a derived from MSCs can inhibit the proliferative, migrating, and invasive abilities of HCC cells by downregulating SALL4 expression. The exosomes successfully deliver miR-15a to HCC cells, leading to decreased tumorigenicity and metastasis of HCC tumors. In vivo experiments in nude mice further confirmed the inhibitory effects of exosomal miR-15a on HCC development.

Summary: Lactate-driven upregulation of CD44 enhances the release of CD44-enriched exosomes in malignant Glioblastoma cells, promoting tumor progression. Sensitive detection and quantification of enhanced CD44 and CD133 levels in immunocaptured exosomes using TiN-NH-LSPR biosensors support its potential application in minimally invasive molecular diagnostics for GBM progression.

Cancer-secreted exosomal miR-1468-5p promotes tumor immune escape via the immunosuppressive of vessels

Chenfei Zhou et al.

Summary: The study suggests that miR-1468-5p in cervical cancer exosomes promotes lymphatic PD-L1 upregulation and lymphangiogenesis, leading to T cell immunity suppression and allowing cancer cells to evade anti-cancer immunity. High levels of exosomal miR-1468-5p in serum correlate with immunosuppressive status in the tumor microenvironment and poor prognosis in cervical cancer patients.

Neutral sphingomyelinase-2 and cardiometabolic diseases

Summary: Sphingolipids, especially ceramides, are crucial in pathophysiological processes related to metabolic syndrome, influencing insulin resistance, type 2 diabetes, and other diseases. Among sphingomyelinases, neutral sphingomyelinase 2 (nSMase2) appears to be particularly important in inflammation-related pathologies, with a feed-forward interaction mechanism involving proinflammatory cytokines. Inhibiting nSMase2 may have therapeutic benefits against inflammation-driven cardiometabolic diseases.

Caveolin-1-driven membrane remodelling regulates hnRNPK-mediated exosomal microRNA sorting in cancer

Harley Robinson et al.

Summary: The study reveals a novel mechanism in which non-caveolar CAV1 drives the loading of metastasis-promoting microRNAs into exosomes by regulating hnRNPK localization, with this process being modulated by the lipid environment of membranes. Knockdown of hnRNPK in prostate cancer cells impairs the ability of extracellular vesicles to induce osteoclastogenesis, indicating a potential role in bone metastasis. Additionally, elevated levels of hnRNPK in biofluid extracellular vesicles are associated with metastatic prostate and colorectal cancer.
