Edaravone ameliorates depressive and anxiety-like behaviors via Sirt1/Nrf2/HO-1/Gpx4 pathway

Summary: Inflammation and oxidative stress have been implicated in the development of depression. This study investigated the antidepressant effects and mechanisms of action of Edaravone (EDA), a free radical scavenger. The results demonstrated that EDA alleviated depressive and anxiety-like behaviors in a chronic social defeat stress (CSDS) depression model. Furthermore, EDA reduced neuronal loss, oxidative stress damage, and activation of inflammatory cytokines in the hippocampus and medial prefrontal cortex. The antidepressant effects of EDA were mediated through the Sirt1/Nrf2/HO-1/Gpx4 signaling pathway and Gpx4-mediated ferroptosis.

Summary: The anti-inflammatory signal of the vagus nerve mediated by alpha 7 nicotinic acetylcholine receptor (alpha 7nAchR) plays a critical role in the antidepressant effect of transcutaneous auricular vagus nerve stimulation (taVNS). The hippocampus region regulates emotions and has the most distribution of alpha 7nAchR, indicating its involvement in the taVNS antidepressant effect.

Metabolomics based comprehensive investigation of Gardeniae Fructus induced hepatotoxicity

Summary: This study investigates the mechanism of hepatotoxicity induced by Gardeniae Fructus (ZZ) using metabolomics both in vivo and in vitro, revealing disruptions in purine and amino acid metabolism in the liver and pyrimidine, primary bile acids, amino acid metabolism, and pantothenate and CoA biosynthesis in the plasma. The study suggests that hepatotoxicity can be identified in cells, providing a non-animal alternative for systemic toxicology research.
