A unifying framework for amyloid-mediated membrane damage: The lipid-chaperone hypothesis

Summary: Researchers have focused on a class of proteins that form pathogenic amyloid aggregates in vivo and their role in various diseases. These proteins, known as intrinsically disordered proteins (IDPs), can partially fold into functional conformations and accumulate as amyloid aggregates upon interaction with other cellular partners. Recent studies have also shown the harmful effects of amyloid proteins on membrane integrity. However, the molecular mechanism underlying the transfer of these proteins from the aqueous environment to the membrane core is still poorly understood.

Electrochemical assessment of dielectric damage to phospholipid bilayers by amyloid β-Oligomers

Summary: This study investigates the damage caused by A beta(1-42) oligomers to tethered lipid membranes (tBLMs) and compares the effects with and without the oligomerization inhibitor HFIP. The results show that membranes containing sphingomyelin are the most susceptible to damage by A beta(1-42) oligomers. The analysis also reveals the formation of highly conductive water-filled nanopores in sphingomyelin membranes by A beta(1-42) oligomers without HFIP.
