The emerging role of the Angiopoietin-Tie pathway as therapeutic target for treating retinal diseases

Lorenzo Ferro Desideri et al.

Summary: Simultaneously targeting the VEGF and Ang/Tie pathways may be more beneficial than monotherapy in patients with exudative retinal diseases. Among the investigational drugs targeting the Ang/Tie pathway, faricimab has shown promising results in phase II/III trials and in the near future may represent a viable treatment option for the management of exudative macular diseases.

Trends in prevalence of blindness and distance and near vision impairment over 30 years: an analysis for the Global Burden of Disease Study

Rupert R. A. Bourne et al.

Summary: The age-adjusted prevalence of blindness has decreased over the past three decades, but the number of people affected has actually increased due to population growth. By 2050, it is predicted that there will be an increase in the number of individuals suffering from vision impairments, highlighting the need for continued efforts in addressing this global issue.

In situ evaluation of spatiotemporal distribution of doxorubicin from Drug-eluting Beads in a tissue mimicking phantom

Marcus Caine et al.

Summary: The study compared a custom version of the traditional phantom sectioning approach with a novel confocal microscopy technique for drug distribution measurements, finding that confocal microscopy enables real-time measurement of drug concentration. Different types of beads exhibited variations in drug concentration and penetration distance. Comparison of in vitro results with historical in vivo data showed good consistency.

Retina stem cells, hopes and obstacles

Olga L. German et al.

Summary: Retinal degeneration is a major cause of visual dysfunction globally. Using stem cells for cell replacement therapy shows great potential in treating this condition. Preclinical research and clinical trials suggest that RPCs and iPSCs are the most promising treatment options, but there are still significant concerns and challenges that need to be addressed.

Two Complementary Signaling Pathways Depict Eukaryotic Chemotaxis: A Mechanochemical Coupling Model

Luewen Zhou et al.

Summary: The study proposes a new cell migration model with a multilayered dynamic seesaw mechanism and a mechanical strain-based inhibition mechanism, demonstrating cell migration and morphological changes through simulations. The delicate balance between endogenous signaling and mechanically governed inactivation scheme is revealed, accounting for the correlated random migration of cells.

Effectiveness of Microcurrent Stimulation in Preserving Retinal Function of Blind Leading Retinal Degeneration and Optic Neuropathy: A Systematic Review

Jinfeng Liu et al.

Summary: Electrical stimulation therapy shows promising therapeutic effects on preserving visual function in retinal degeneration and optic neuropathy. More large-scale randomized controlled trials are needed to explore its potential in age-related macular degeneration, retinal artery occlusion, and glaucoma. The lack of comparison between different ES methods in the same disease populations highlights the need for further research to optimize parameters and detection methods.

Intraocular pressure and injection forces during intravitreal injection into enucleated porcine eyes

Andrea Allmendinger et al.

Summary: This study investigated the effect of injection volume on intraocular pressure using healthy enucleated porcine eyes, revealing an exponential increase in IOP for injection volumes larger than 100 µL. Interestingly, there were no significant differences in IOP based on viscosity, injection rate, and needle diameter, but variability increased significantly for injection volumes larger than 100 µL. Additionally, the exponential increase in IOP was not reflected by injection force measurements for typical configurations used for intravitreal injection, providing important insights for injection volume guidance and injection force considerations in drug product development.

Human stem cell-based retina on chip as new translational model for validation of AAV retinal gene therapy vectors

Kevin Achberger et al.

Summary: Gene therapies using AAVs show promise in treating or curing retinal diseases, but the development of efficient AAV vectors is slow and costly due to the lack of suitable non-clinical models. Human iPSC-derived retinal organoids faithfully recreate human tissue structure and function, making them essential for translational research. OoC technology allows for the recapitulation of tissue environments and precise control over parameters, with iPSC-based OoC models showing potential as the next generation of screening platforms for gene therapeutic studies.
