The cytokine storm and COVID-19

Biying Hu et al.

Summary: COVID-19, caused by SARS-CoV-2, has become a global pandemic since its emergence in Wuhan, China, in December 2019, posing a serious threat to public health. Close contact and respiratory droplets are the main modes of transmission. The cytokine storm is closely associated with disease severity and mortality, emphasizing the importance of studying and treating the cytokine storm in COVID-19.

Clinical studies assessing the efficacy, effectiveness and safety of remdesivir in management of COVID-19: A scoping review

Juan Pimentel et al.

Summary: Remdesivir may shorten recovery time and improve clinical status in severe COVID-19 patients, with evidence suggesting decreased mortality and increased survival. However, the most common adverse reaction is hepatic impairment, although a similar proportion of adverse events is reported in treated and control groups. Results from ongoing trials are expected to provide more insights into the efficacy and safety of this antiviral drug, with further clinical trial results in 2020 allowing for a quantitative synthesis of available evidence.

Population Pharmacokinetic Evaluation of Amikacin Liposome Inhalation Suspension in Patients with Treatment-Refractory Nontuberculous Mycobacterial Lung Disease

Christopher M. Rubino et al.

Summary: The study developed a population pharmacokinetic model to evaluate the pharmacokinetic properties of once-daily amikacin liposome inhalation suspension (ALIS) in patients with treatment-refractory NTM lung disease. Results showed that systemic exposure to amikacin in serum and urine following once-daily ALIS administration was notably lower than that previously reported for parenteral amikacin.

SARS-CoV-2 M-pro inhibitors with antiviral activity in a transgenic mouse model

Jingxin Qiao et al.

Summary: The study designed and synthesized 32 new M-pro inhibitors containing bicycloproline, which showed inhibitory effects on SARS-CoV-2. Compounds MI-09 and MI-30 exhibited excellent antiviral activity in cell-based assays and significantly reduced lung viral loads and lung lesions in a transgenic mouse model of SARS-CoV-2 infection. Both also displayed good pharmacokinetic properties and safety in rats.

Safety, tolerability, and immunogenicity of an aerosolised adenovirus type-5 vector-based COVID-19 vaccine (Ad5-nCoV) in adults: preliminary report of an open-label and randomised phase 1 clinical trial

Shipo Wu et al.

Summary: The study evaluated the safety and immunogenicity of an aerosolised adenovirus type-5 vector-based COVID-19 vaccine in adults without COVID-19 from China. It found that the aerosol vaccine was well tolerated, and two doses elicited neutralising antibody responses similar to one dose of intramuscular injection. An aerosolised booster vaccination at 28 days after the first intramuscular injection induced strong IgG and neutralising antibody responses. The efficacy and cost-effectiveness of aerosol vaccination should be evaluated in future studies.

Structural analysis of carbohydrate binding by the macrophage mannose receptor CD206

Hadar Feinberg et al.

Summary: The human mannose receptor expressed on macrophages and hepatic endothelial cells plays an important role in reducing tissue damage by scavenging released lysosomal enzymes, glycopeptide fragments of collagen, and pathogenic microorganisms. By binding to mannose, fucose, or N-acetylglucosamine (GlcNAc) residues on these targets, the receptor interacts with sugars in a Ca2+-dependent manner through its C-type carbohydrate-recognition domain 4 (CRD4), which has been investigated through glycan array screening and multiple crystal structures. The interactions with monosaccharides and oligosaccharide substructures present in these ligands provide insight into the mechanisms of binding and potential classes of ligands for the mannose receptor.
