Two types of O-methyltransferase are involved in biosynthesis of anticancer methoxylated 4′-deoxyflavones in Scutellaria baicalensis Georgi

Meng-Ying Cui et al.

Summary: The study found that methoxylated flavones have strong ability to induce apoptosis and enhance bioactivity, with methoxylation not causing side effects on healthy cells. Both type I and type II OMT enzymes are involved in the biosynthesis of methoxylated flavones in S. baicalensis.

Differential transcriptome analysis of genes associated with the rhizome growth and sesquiterpene biosynthesis in Atractylodes macrocephala

Qingyan Ruan et al.

Summary: This study conducted metabolic analysis and transcriptome sequencing of different tissues of Atractylodes macrocephala, identifying key transcription factors involved in regulating rhizome growth and sesquiterpene biosynthesis. EF-1α was found to be the best reference gene for tissue expression pattern analysis. The extensive transcriptome data provided a valuable resource for further research on molecular mechanisms of rhizome growth and sesquiterpene biosynthesis.

Integration of full-length transcriptomics and targeted metabolomics to identify benzylisoquinoline alkaloid biosynthetic genes in Corydalis yanhusuo

Dingqiao Xu et al.

Summary: This study identified candidate genes involved in the biosynthesis of benzylisoquinoline alkaloids (BIAs) in Corydalis yanhusuo through full-length transcriptomic and metabolomic analyses. A complete BIA biosynthesis pathway was constructed based on the findings, and phylogenetic analysis revealed enzymes with columbamine-O-methyltransferase activity. The results provide a foundation for further molecular cloning of enzymes and activity validation studies.

Erythrina velutina Willd. alkaloids: Piecing biosynthesis together from transcriptome analysis and metabolite profiling of seeds and leaves

Daisy Sotero Chacon et al.

Summary: This study presents a novel approach to analyze a non-model Fabaceae species through combined transcriptome and metabolome analyses. The results revealed the presence of 42 alkaloids in seeds and leaves of E. velutina, some of which are unique. By identifying 24 gene candidates, a putative pathway leading to the singular alkaloid diversity of this species was proposed, indicating potential biotechnological targets for modulation of erythrina alkaloids biosynthesis.

Multi-omics analysis of the bioactive constituents biosynthesis of glandular trichome in Perilla frutescens

Peina Zhou et al.

Summary: Perilla frutescens (L.) Britt is a widely cultivated medicinal and edible plant in Asia, with terpenoids, flavonoids, and phenolic acids as the main medicinal ingredients. Glandular trichomes with multicellular structures are considered biochemical cell factories in which specialized metabolites are synthesized. This study explored the morphological features of glandular trichomes, metabolic profiling, and transcriptomes in different tissues of P. frutescens.

CRISPR/Cas9-mediated targeted mutagenesis of bZIP2 in Salvia miltiorrhiza leads to promoted phenolic acid biosynthesis

Min Shi et al.

Summary: The study identified bZIP2 as a negative regulator in phenolic acid biosynthesis, which can reduce phenolic acids production by suppressing the expression of the PAL gene. Additionally, bZIP2 physically interacts with multiple proteins, providing new insights for metabolic engineering of phenolic acids.

An extremely promiscuous terpenoid synthase from the Lamiaceae plant Colquhounia coccinea var. mollis catalyzes the formation of sester-/di-/sesqui-/mono-terpenoids

De-Sen Li et al.

Summary: This study identified a highly promiscuous terpenoid synthase, CcTPS1, which is the first enzyme in the plant kingdom capable of generating a variety of terpenoids. CcTPS1 exhibits leaf-specific expression and is mainly responsible for the biosynthesis of diterpenoids and sesquiterpenoids. The findings contribute to our understanding of plant terpenoid synthase evolution and natural product diversity.
