Confirmation of botulism diagnosis in Australian bird samples by ELISA and RT rtPCR

Anne M. Masters et al.

Summary: A sandwich ELISA and RT-rtPCR were developed to detect Clostridium botulinum C and D toxins, replacing the mouse bioassay. The diagnosis of botulism in birds was confirmed through sample testing, with the RT-rtPCR for the botulinum C toxin gene proving to be helpful in confirming the diagnosis.

Modification and validation of the Endopep-mass spectrometry method for botulinum neurotoxin detection in liver samples with application to samples collected during animal botulism outbreaks

Annica Tevell Aberg et al.

Summary: The Endopep-MS method has been successfully modified and validated for analyzing cattle, horse, and avian liver samples, with the introduction of a combination of a salt washing step and a protease inhibitor cocktail. These modifications resulted in a substantial decrease in interfering signals and an increase in BoNT-specific signals, leading to a substantial improvement in sensitivity, especially for BoNT-C and C/D which are among the most prominent serotypes for animal botulism. Botulism was diagnosed with the new method in liver samples from dead cattle and birds from outbreaks in Sweden.
